Objective:Gastric cancer is one of the major malignant tumors that are harmful to human health.It is one of the most common malignant tumors in the world.Because of the lack of effective means of early diagnosis of gastric cancer,the vast majority of gastric cancer in clinical diagnosis has been advanced,resulting in high mortality.At present,the treatment of gastric cancer is mainly surgery,and the application of neoadjuvant chemotherapy can increase the survival rate of five years,but the overall effect is still not ideal.With the development of molecular biology,the early diagnosis and molecular targeted therapy of gastric cancer have become the focus of research.In recent years,A class of molecules known as long chain non coding has been paid more and more attention because of its important role in post transcriptional regulation,cell growth,differentiation and proliferation.The relationship between long chain noncoding RNA and tumorigenesis has become the focus of research To investigate the expressions of MEG3,GAS5 in gastric cancer tissues and and their relationships with the prognosis,and to explore the relationships between MEG3 and apoptosis.Methods:From September 2012 to June2013,55 consecutive Patients with gastric cancer admitted to Qingdao Municipal Hospital for surgical treatment were included into our study.We collect gastric cancer tissues and paired normal tissues.We tested the relative expression of MEG3,GAS5 genes by real-time quantitative PCR(RT-PCR),and We tested the expression of P53 and MDM2 in gastric cancer by Western blot and to evaluate their correlations with MEG3.Results:1.The expression of lnc RNA MEG3 was decreased in cancer tissues(7.98±0.19)relative to normal tissues(9.47±0.18)(P<0.05).2.The expression of lnc RNA GAS5 was decreased in cancer tissues(4.03±0.18)relative to normal tissues(6.06±0.18),(P<0.05).3.The expression of MEG3 and GAS5 correlated with the degree of differentiation,lymph node metastasis and TNM stage.4.It showed that there were positive relationships between P53 and MEG3(r=0.591,P<0.05),and there were negative relationships between MDM2 and MEG3(r=-0.346,P<0.05).5.The median survival time was significantly prolongged in the patients whose expression of MEG3 and GAS5 were high than the low.Conclusions: 1.MEG3 and GAS5 play inhibiting effects on the cancer gene;2.MEG3 and P53,MDM2 in the pathogenesis and the development of biological mechanism of gastric cancershave important links3.To detect the expression levels of MEG3 and GAS5 have a potential value for the prognosis of gastric cancer.
