Objective:To compare the different expression of long non-coding RNAs in Her2(+)gastric cancer tissues and Her2(-)gastric cancer tissues,to find long non-coding RNAs that are differentially expressed in Her2(+)gastric cancers,and to investigate the impact of long non-coding RNAs on the development and progression of Her2(+)gastric cancer.Methods:Collected 5 cases of Her2(+)gastric cancer tissue specimens and 5cases Her2(-)gastric cancer tissue specimens that surgically resected from General Hospital of Tianjin Medical University.Real-time fluorescence quantitative polymerase chain reaction(RT-qPCR)was used to detect the differential expression of some long-chain non-coding RNAs in two types of gastric cancer.Using purified PCR product of target genes to prepare standard curves and to detect the different expression of TUG1,DANCR,CRNDE,PVT1,and MIR31HG in Her2(+)and Her2(-)gastric cancer tissues by RT-qPCR.Result:1.The expression of LncRNA TUG1(8.84×10~4±2.88×10~4 copies/ng)was higher in Her2(+)gastric cancer tissue than in Her(-)gastric cancer tissues(3.08×10~4±1.89×10~4copies/ng),and the difference was significant(p=0.006).2.The expression of LncRNA PVT1(325.94±174.02copies/ng)in Her2(+)gastric cancer tissue samples was lower than that in Her2(-)gastric cancer tissues(993.1±398.1 copies/ng),and the difference was significant(p=0.009).3.DNACR,CRNDE,MIR31HG,and these three long non-coding RNAs were insignificantly different in Her2(+)gastric cancer tissues and Her2(-)gastric cancer tissues(p>0.05).Conclusion:1.There exists differentially expressed LncRNA between Her2(+)gastric cancer tissue and Her2(-)gastric cancer tissue.2.The expression of LncRNA TUG1 in Her2(+)gastric cancer tissues was higher than that in Her2(-)gastric cancer tissues.The expression of LncRNA PVT1 in Her2(+)gastric cancer tissues was lower than that in Her2(-)gastric cancer tissues.These two LncRNA may affect the occurrence and development of Her2(+)gastric cancer by affecting the expression of p21 and p53 proteins.3.There was no significant difference in the expression of DANCR,CRNDE and MIR31HG between Her2(+)gastric cancer tissues and Her2(-)gastric cancer tissues.They may not participate in the occurrence and development of Her2(+)gastric cancer.
