In this study,the author screened out the natural drugs with the function of resisting type 2 diabetes with hyperuricemia.Ethanol was extracted from 86 medicinal plants,inhibition of alpha glucosidase in ethanol extracts of medicinal plants in vitro,inhibition of xanthine oxidase activity were screened,and their good activity of medicinal plant extracts were screened in vivo blood sugar and blood uric acid activity,of selected medicinal plants ethanol extracts were analyzed determination of total flavonoids content and infrared spectroscopy.The main research contents and results were as follows:(1)The ethanol extracts of 86 medicinal plants were prepared and glucose oxidase method was used to determine the absorbance value(A)of the 86 kinds of medicinal plants,and the inhibition rate of the alpha glucosidase was determined by using the microplate reader at nm 490.Alcohol extract of the torch tree,a.triflorum,Ligularia jaluensis,Acer ginnala,erigeronannus ground,phtheirospermum,Rhodiola rosea,Huaihua,epimedium,Ligustrum lucidum and Rhizoma Paridis was screened out.The inhibition rate reached more than 80% with their concentration of 20 mg· mL-1.(2)For xanthine oxidase inhibitory activity screening of 86 medicinal plants ethanol extracts by ELISA method in 2 mmol-L-1 inhibition rate of more than 60% screening,according to the average inhibition rate of each sample IC50,determine the type of inhibition kinetics of xanthine oxidase was analyzed.There are 10 species of medicinal plant extracts of Ligularia jaluensis,Huaihua,serviceberry alcohol torch leaves,a.triflorum,epimedium,blue honeysuckle,Rhodiola rosea,Xunyang double coneflower and Geum alcohol extracts are very effective in vitro xanthine oxidase inhibition.The 10 kinds of medicinal plants ethanol extracts were analyzed for determination of total flavonoids content and infrared spectroscopy,to determine those active ingredients.(3)Research of hypoglycemic activity of mouse of STZ induced type 2 diabetes was done.The results showed that the torch tree,a.triflorum,Ligularia jaluensis,erigeronannus ground,phtheirospermum and sophoranol extracts could effectively reduce blood glucose levels in diabetic mice,but by the determination of serum indicators in diabetic mice,the torch tree,Acer triflorum and Huaihua were best.(4)To establish hyperuricemia model mice induced by yeast extract,10 species of medicinal plants were screened in vitro for alcohol extract in vivo uric acid lowering activity were further screened,and the detection of serum indicators and the kidney pathology observation.The torch tree,Acer triflorum,Rhodiola rosea and sophoranol extract in detection of uric acid and serum index showed strong activity to restore kidney atrophy phenomenon better.(5)The screened respectively to blood glucose and blood uric acid of the medicinal plant extracts,further research is torch tree,a.triflorum and sophoranol extract against type 2 diabetes mellitus and hyperuricemia.With the establishment of yeast extract in mice induced by type 2 diabetes mellitus and hyperuricemia model by using STZ,results show that the content of uric acid and blood glucose levels in mice model group were higher than the normal group,and renal tissue damage is serious,can determine the successful model,three kinds of medicinal plants ethanol extracts on type 2 diabetes with hyperuricemia model mice blood glucose and blood uric acid could decrease,and has a positive effect on the content of serum index,can restore the phenomenon of renal tissue damage.Through the screening of 86 medicinal plant extracts against type 2 diabetes mellitus and hyperuricemia activity,screened from three species of medicinal plants were reported,and the torch tree,a.triflorum and Huaihua,in the course of the study,some indicators may not be ideal,but still has a tendency and dose-dependent manner.
