In the deep culture of human history,wine is not only an objective material existence but also a symbol of culture.As the development of social progress and economy,wine has been a necessity of family meeting and social situation.In the meanwhile,with the increasing of acute alcohol intoxication and poisoning caused by drinking in excess,it also has impacted human’s health.Therefore,development and research of safe,effective antialcoholic products become the hot topic in the whole world.Nowsdays,the domestic research plays more attention to the original extract from traditional Chinese medicine.However,if these medicine be used for long time,it will harm human’s body.thus,the preventive therapy of diet always be the hot spot for research.And the plant polysaccharide is unique in this respect.It is significant that develop vegetal polysaccharides functional foods for preventing injury from alcohol.Konjac in the Ministry of Health announced the "medicine and food dual purpose" list of items is known as ordinary food management of new food resources.At present,it has been reported that the konjac glucomannan has the effect of anti-inebriation and protective effect of alcoholic liver injury,some scholars believe that konjac has the physical adsorption of ethanol so as to prevent drunk hangover,but these views just stay in the hypothetical level and lack of experimental confirmation,so konjac hangover function still need further systematic study.In this paper,we explore konjac hangover from the two aspects of in vitro and in vivo by dialysis bag method simulated gastrointestinal environment in vitro,the effects of Konjac polysaccharide on the precipitation of ethanol from the dialysis bag were studied under various factors,the physical adsorption of konjac to ethanol was also studied.By means of feeding in different doses of konjac powder(166,332,500 mg/kg),the Kunming mice as test subjects,using 56 ° Hongxing Erguotou gavage methods of modeling,exploring the effect of konjac on the enhancement of first-pass metabolism and the protective effect on the gastric mucosa and the antioxidant damage to the brain and the regulation of neurotransmitters in acute alcohol mice in vivo,in order to provide the theory andsupport for the development of vegetal polysaccharides functional food for the prevention and treatment of alcoholic injury.The main conclusions of this paper are as follows:1.The research was on the change of ethanol precipitation and the physical adsorption effect of konjac flour under different factors in simulated gastrointestinal environment in vitro.A.in the simulated gastric juice and simulated intestinal fluid,taking time,ethanol concentration and KGM concentration as experimental factors,the KGM-ethanol mixture was added into the dialysis bag,the water-ethanol mixture was used as the control group,determining the changes of ethanol content in the control group and the konjac flour group under different factors by gas chromatography,the results showed:(1)in different time periods,comparing with control group,KGM group was significantly lower about ethanol dialysis volume in the period of 20-90 min,but no significant difference with the control group in the period of 120-210 min in simulated gastric fluid,there was significant differences in the amount of ethanol in the KGM group in intestinal juice in each time,indicating that KGM could reduce the amount of free ethanol,which may play a certain role to reduce and delay the ethanol into the blood stream,and with the extension of time,the amount of ethanol dialysis in the KGM group gradually increased,that indicating the KGM could prevent the permeation of ethanol in a short time,and with the extension of time,the ethanol concentration on both sides of the dialysis bag reached a dynamic equilibrium at 150 min.(2)Under the same KGM concentration and different ethanol concentration,ethanol dialysis of KGM group was obviously lower control group,with the increase of ethanol concentration in the range of 10%-40%(v/v),the amount of ethanol precipitation gradually increased and showed significant difference,with the increase of ethanol concentration in the range of 40%-60%(v/v),there was no significant difference in the amount of ethanol in the KGM group,indicating that the effect of KGM in high concentration ethanol was stronger than that of low concentration ethanol.(3)Under the same ethanol concentration and different KGM concentration,the ethanol content of the KGM group was significantly lower than that of the control group,at the same time,the amount of ethanol precipitation decreased than low concentration of KGM in high concentration,which indicated that the adsorption capacity of ethanol increased with the increase of KGM concentration.B.In the test of KGM adsorption of ethanol,using water-ethanol mixture as control group,by the mixing and centrifugation of the konjac colloid and the different concentrations of ethanol solution,compared with the control group,theethanol content of the supernatant from the KGM group was decreased by the mixing and centrifugation of the konjac colloid and the different concentrations of ethanol solution,indicating that KGM had an adsorption effect on ethanol,reducing the absorption of alcohol in the digestive tract through the physical,reducing alcohol content in blood,avoiding disposable alcohol poisoning caused by drinking too much,so as to achieve the purpose of preventing drunkenness.By measuring the adsorption rate of a certain amount of KGM at different ethanol concentrations,with the ethanol concentration in the range of 10%-50% increase,the adsorption of KGM on ethanol decreased but no significant,when the ethanol concentration reached 60%(v/v),compared with the KGM adsorption rate of other ethanol concentration range,the adsorption rate of KGM on ethanol decreased and there was significant difference,indicating that a certain amount of KGM can not be unlimited adsorption of ethanol,the ability could be limited after reaching a certain amount of ethanol.2.Enhancement of konjac powder on the metabolism of ethanol and its protective effect on gastric mucosa,in vitro experiments,konjac powder could reduce blood alcohol concentration by adsorption to ethanol,when the intake of ethanol is too large,it is mainly enhance the first pass metabolism to reduce the amount of free ethanol in gastrointestinal fluid.The results showed: compared with the blank group,the activity of ADH and ALDH in the gastric tissue homogenate of the model group decreased and the ethanol concentration in the blood increased.Compared with the model group,the activity of ADH and ALDH significantly increased and the concentration of blood alcohol decreased in the konjac flour group,indicating that konjac flour could enhance activity of ADH and ALDH in the stomach,promote the first metabolism and inhibit the absorption of gastrointestinal to ethanol,reduce concentration of blood alcohol.Comparing with the blank group,stomach edema index,gastric mucosa ulcer index increased significantly,free acid and total acid showed an upward trend in addition to pH value had no significant differences in gastric acid,TNF-α content increased,the SS levels drop in biochemical indexes,gastric mucosa pathological tissue damage was serious,suggesting that modeling success.low,middle and high dose groups of konjac powder were significantly different from those in the model group in content of TNF-α,while in SS level,there was difference in the low dose group but not significant,there was significant difference between the middle and high dose groups,indicating that konjac powder can protect the gastric mucosa by regulating the production of inflammatory mediators.3.The research was on antioxidant protection of Konjac powder on acute alcoholism brain injury.The results showed that: the contents of MDA and NO in the brain tissue of the model group were significantly higher than those in the blank group,and the contents of SOD and GSH were significantly decreased in the model group,observating on pathological section in brain tissue,the number of hippocampal pyramidal cells decreased,arranged in loose,and the cells were necrotic and atrophic in some areas in the model group,whole cells of konjac powder group closely packed,the number of cells is more than model group.The mice were administered with different doses of konjac powder,the level of SOD and GSH in the low and middle dose groups of the konjac powder was higher than that of the model group,but there was significant difference between the two groups,however,there was significant difference between the high dose group and the model dose group,the content of MDA in middle and high dose group was significantly lower than that in model group,high dose group compared with model group increased in NO levels and is significant difference.On the whole,the antifungal capacity of konjac powder high dose group was better than that of low and middle dose group,and it could regulate metabolism of the free radical in brain tissue after drinking and avoid oxidative damage and play a protective role in brain tissue of mice with acute alcoholism.4.The research was on the effect of Konjac powder on acute alcoholism acetaldehyde and brain neurotransmitter regulation,the results showed that compared with the blank group,the content of acetaldehyde in the blood and brain of mice in the model group,the content of β-EP,DA,5-HT in brain increased significantly,and these indexes were significantly different.The content of β-EP in each group of konjac flour decreased and there was significant difference.The content of DA and 5-HT were significantly different in the middle and high dose group of konjac flour,while the low dose group of konjac flour was lower but no significant.And on the whole,the effect of high dose group was better than that of konjac powder of konjac powder in low and medium dose group.
