Objective:The purpose of this study is to verify the biocompatibility of carbon fiber composites with osteoinductive activity in acute phase.Methods:1.Cytotoxicity test:The cells are cultured with the extract of the material,the growth of the cells is observed,and the absorbance value of the cells is measured by CCK-8Kit,and the relative proliferation rate of cells was calculated;2.Acute toxicity test: The activity of ICR mice is observed by intraperitoneal injection of the extract;3.Micronucleus test: The micronucleus rate of bone marrow of ICR mice is detected by intraperitoneal injection of the extract;4.Chromosome aberration test:The frequency of chromosome aberration of ICR mice is detected by intraperitoneal injection of the extract and colchicine treatment;5.Intradermal stimulation test: The extract is injected subcutaneously into the back of the New Zealand white rabbits,and the rabbit skin response is observed;6.Pyrogen test: The extract is injected into New Zealand white rabbits through the ear vein,and then the body temperature is observed;7.Hemolysis test:The extract is added to the anticoagulant blood of New Zealand white rabbits,and then the hemolysis is observed and the absorbance is measured;8.Sensitization test: The extract is injected subcutaneously into the back of guinea pig to induce sensitization,and then sensitize in the ventral side.Finally,the appearance and pathological changes of the skin are observed.Results:1.Cytotoxicity test:The cells were elongated,and the growth of the cells was good,and the relative increment rates of A,B and C groups were 75% at different time;2.Acute toxicity test:The mice of A,B and C groups had good activity and no adverse reaction;3.Micronucleus test:The micronucleus rates of A,B and C groups were1.40‰,1.80 ‰ and 1.20‰;4.Chromosome aberration test:There was no chromosome aberration in A,B and C groups;5.Intradermal stimulation test:The average primary irritation index of A,B and C groups were less than 0.4;6.Pyrogen test:The temperature rise values of A,B and C groups were lower than 0.6?,and the total values were below1.4 ?;7.Hemolysis test: Hemolysis rates of A,B and C groups were less than5%;8.Sensitization test: The sensitization rates of A,B and C groups were 0,and the pathological sections showed no abnormality.Conclusion:Carbon fiber composite material with osteoinductive activity for repairing skull defects has no cell toxicity,no acute toxicity,no genetic toxicity,no irritation to the skin,will not cause the increase in body temperature,no sensitization,and conforms to the standard of clinical application of medical materials.
