Objective:In experimental study,THP-1 cell-derived foam cells were cultured by HDL extracted in normal healthy people and smoking patients with coronary heart disease.The effects of HDL on the reversal of cholesterol in THP-1 cell-derived foam cells were observed.Promote the pathogenesis of atherosclerosis to provide a theoretical basis.Methods:Blood samples were collected from normal healthy people and people with coronary heart disease,and high density lipoprotein was extracted by ultra high speed centrifuge.THP-1 cells were induced to be macrophages by phorbol ester and then induced by oxidized low density lipoprotein(ox-LDL)The cells were divided into three groups: blank control group,normal human HDL group and smoking group.By Oil Red O staining was observed in the group foam cell lipid droplets case;using a liquid scintillation counter to detect the respective cholesterol efflux rate of cells in each group from the apparent phenomena;Group 3 adenosine triphosphate using Western blot detection more binding cassette transporter A1(ABCA1),Adenosine triphosphate conjugate transporter G1(ABCG1),B classⅠscavenger receptor(SR-BI)corresponding protein content.Results:⑴successfully extracted human high-density lipoprotein,and identification;⑵PMA successfully induced THP-1 cells to form foam cells,ox-LDL successfully induced foam cell formation;(3)Results compared with the control group,normal human HDL group and smoking coronary heart disease group can make the cholesterol efflux rate increased(P<0.05),normal human HDL group has a more significant effect compared with smoking coronary heart disease group(P<0.05);⑷Compared with the blank control group,the expression of ABCA1 and ABCG1 protein normal human HDL group had no significantchange(P>0.05),and the expression of ABCA1 and ABCG1 protein in smoking coronary heart disease group was significantly decreased(P <0.05);⑸Compared with blank control group,the expression of SR-B1 protein in normal human HDL group and smoking coronary heart disease group was significantly increased(P <0.05).The expression of SR-B1 protein in normal human HDL group was significantly higher than that in and smoking coronary heart disease group(P <0.05).Conclusion:Normal HDL can promote the reverse transport of cholesterol in THP-1 cell-derived foam cells,promote cholesterol efflux,and it can increase the expression of SR-B1 protein,but it does not affect ABCA1,ABCG1 protein expression,and smoking coronary heart disease HDL compared to normal HDL,promote cholesterol efflux and increase the expression of SR-B1 protein weakened,and can significantly reduce the ABCA1,ABCG1 protein expression.To provide a theoretical basis for the promotion of the pathogenesis of atherosclerosis,so as to reduce or avoid the occurrence and development of AS,and provide a theoretical basis for clinical prevention and treatment of coronary atherosclerotic heart disease.
