Objective:To investigate the correlation between UCP3-55C/T polymorphism and fasting plasma glucose(FBG),fasting insulin level(FINS),insulin resistance(HOMA-IR)and type 2 diabetes mellitus.Methods:A total of 108 newly diagnosed type 2 diabetes mellitus patients(58 males and 50 females)and 110 non-diabetic patients(52 males and 58 females)with UCP3-55C?T were detected by Snapshot typing.Measure the height(cm),body weight(kg),waist circumference(cm),hip circumference(cm)respectively.Calculate body mass index(BMI = body weight(kg)/ square of height),waist to hip ratio(WHR = waist circumference / hip circumference).Glucose oxidase method was used to detect fasting plasma glucose and enzymatic determination of fasting triglyceride(TG)and total cholesterol(TC).High density lipoprotein cholesterol(HDL-C)and low density lipoprotein cholesterol(LDL-C)were detected by dextran sulfate-manganese precipitation method and calculated using the formula respectively.Radiofrequency immunoassay was used to detect fasting blood insulin(FINS).Steady-state model assessment(HOMA-IR)was used to calculate the insulin resistance index [FINS × FPG / 22.5].Results:(1).The frequency of genotype distribution of UCP3 promoter-55C/T polymorphism in non-diabetic group was CC 38.18%(42),CT 45.46%(50),TT 16.36%(18),C and T allele frequencies were 60.91%,39.09%.The frequency of genotype distribution in type 2 diabetes mellitus was CC 49.08%(53),CT 42.59%(46),TT 8.33%(9),C and T allele frequencies were 70.37% and 29.63%,respectively.Compared with non-diabetic group,the frequency of T allele in type 2 diabetes mellitus was significantly lower than that in non-diabetic group(P<0.05).(2).The FBG,FINS and HOMA-IR of T-gene carriers in the non-diabetic group were significantly lower than those of CC genotype(P <0.05).The FINS and HOMA-IR of T-gene carriers in type 2 diabetes mellitus were lower than those in control group(P <0.05).(3).The levels of BMI and WHR were significantly decreased in the non-diabetic group,and the TG and LDL-C were significantly higher(P <0.05).There were significant differences in BMI,WHR,TG and LDL-C,and HDL-C in the type 2 diabetes mellitus group(P <0.05)).Conclusion:UCP3-55C/T polymorphism was associated with the risk of type 2 diabetes,insulin resistance and FINS in China.T carriers have a lower risk of type 2 diabetes,lower levels of insulin resistance and FINS,This mutation may be a protective factor for type 2 diabetes.At the same time,T carries of UCP3-55C/T have a lower level of BMI,WHR,and a lower risk of obesity.
