Objective:1.To explore the association between-55C/T polymorphism of UCP3 and fasting blood glucose(FBG),glycosylated hemoglobin(HbA1c),insulin resistance(HOMA-IR)in patients with type 2 diabetes.2.To explore the association between-55C/T polymorphism of UCP3 and serum Adiponectin,TNF-? and IL-6 in type 2 diabetes mellitus.Methods:A total of 103 patients(60 males and 43 females)diagnosed with T2 DM were selected as the T2 DM group,and A total of 110 healthy subjects(52 males and 58females)were selected as the normal controls(NC).Polymerase chain reaction-sequencing based typing method was used to determine the genotypes of UCP3-55C?T.The height,weight,waist circumference and blood pressure of the subjects in the two groups were measured and recorded,and the body mass index was calculated.Fasting triglyceride(TG)and total cholesterol(TC)were determined by enzymatic method.High-density lipoprotein cholesterol(HDL-C)and low-density lipoprotein cholesterol(LDL-C)were calculated by dextran sulfate-manganese precipitation method and formula respectively.Fasting plasma glucose(FBG)was detected by glucose oxidase and HbA1 c by HPLC.Fasting blood insulin(FINS)was detected by chemiluminescence.The degree of insulin resistance was assessed by the homeostatic model assessment of insulin resistance index(HOMA-IR = FINS × FPG / 22.5).Adiponectin,TNF-? and IL-6were detected by ELISA.Results:(1)The distribution of UCP3-55 C/T genotype and allele frequency showed no significant difference between NC group and T2 DM group(P = 0.748,0.436,respectively).(2)In T2 DM group,FBG of TT genotype was lower than that of CC genotype,but the difference was not statistically significant.In T2 DM group and NC group,HbA1 c was lower in TT than in CC patients,but the difference was not statistically significant.In the T2 DM group and the NC group,the difference of HOMA-IR among the three genotypes was not statistically significant.(3)The T2 DM patients were divided into three groups according to HbA1c(< 7%,7% ? HbA1 c < 9%,HbA1 c ? 9%).There were significant differences in genotype frequency distribution among three groups(P = 0.024).The proportion of TT genotype in the patients with HbA1 c ? 9% was significantly lower than that of CC genotype(P = 0.043).(4)Spearman correlation analysis indicated that UCP3-55C/T genotype was positively correlated with adiponectin level in T2 DM group(r = 0.256,P = 0.026)and negatively correlated with TNF-? level(r =-0.135,P = 0.038).Compared with CC genotype patients,serum adiponectin level was increased and TNF-? level was decreased in TT and CT genotype patients.Conclusion:Compared with CC patients,HbA1 c of TT patients showed a downward trend.The proportion of TT genotype was significantly reduced in T2 DM patients with poor glycemic control,suggesting that-55C/T of UCP3 polymorphism was related to the level of glycemic control in T2 DM patients.T alleles may improve insulin resistance by increasing serum adiponectin expression and decreasing serum TNF-? expression,thereby improving glycemic control in diabetic patients.
