It is not only as one of the most commonly herbs on Angelicae sinensis Radix used clinical treatment,but also as a major export of TCM varieties to the wold.But the early binding is a serious problem in its actual production,broadcast technology is considered as one of important ways to solve this problem,but it has not to do in-depth research about the quality of the direct seeding Angelicae Sinensis,and not to compare the quality of direct seeding and transplanting Angelicae Sinensis.Therefore,it will make a comparative study on the quality of direct seeding and transplanting Angelicae Sinensis from yield,appearance characteristics,ingredients and pharmacological effects in this paper,the following some contents were systematically researched:A The comparative study on yield and appearance characteristics of direct seeding and transplanting Angelicae SinensisThe weight and appearance characteristics were detected by the conventional method.The results show that the yied of direct seeding Angelicae Sinensis(1576.5kg/mu)2 times higher than transplanting Angelicae Sinensis(679.78 kg/mu),thier appearance is all cylindrical,and the lower of herb have some rootlets,and the surface of herb has some vertical lines and horizontal long lenticels.Looking at the overall from the appearance of herb,the difference of appearance characteristics is smaller between direct seeding and transplanting Angelicae Sinensis,the short growth period may be the cause of small herb and low weight of direct seeding Angelicae Sinensis,but the enough quantity of herbs can make sure the area yield of it,and even higher than that of transplanting Angelicae Sinensis.what's more,less lateral root number of direct seeding Angelicae Sinensis is relatively conforms the characteristics of“straightroot plants” of Angelicae Sinensis.B The comparative study on the contents of index ingredients and other ingredients of direct seeding and transplanting Angelicae SinensisThe contents of total ash,acid-insoluble ash,extract and volatile oil of direct seeding and transplanting Angelicae Sinensis through “Chinese Pharmacopeia 2015”,and ferulic acid was ultrasonic extraction by 70% Methanol-water,and was detected by HPLC.Thier contentsranges of direct seeding Angelicae Sinensis is respectively 4.39% ~ 5.06%,0.42% ~ 0.56%,62.90% ~ 64.80%,0.53% ~ 0.65%,and the average contents of them is respectively 4.85%,0.48%,63.93%,0.57%,the average content of ferulic acid is 0.54 mg/g.however,thier contents ranges of transplanting Angelicae Sinensis is respectively 5.32% ~ 5.46%,0.58% ~0.67%,59.66% ~ 60.94%,0.43% ~ 0.46%,and the average contents of them is respectively5.40%,0.62%,60.41%,0.44%,the average content of ferulic acid is 1.34 mg/g.the contents of index ingredients of direct seeding and transplanting Angelicae Sinensis is all conform to the requirements of the pharmacopoeia,in addition to ferulic acid,the contents of other index ingredients of direct seeding Angelicae Sinensis are better than transplanting Angelicae Sinensis.The method that for ferulic acid was used to detect the contents of Z-ligustilide,3-butylidenephthalide and n-butylphthalide,the amino acids in Angelicae Sinensis were extracted by 0.1 mol/L Hydrochloric acid,and the contents of them were detected by HPLC,the contents of polysaccharides of Angelicae Sinensis were detected by ultraviolet spectrophotometer(UV),and gas chromatography-mass spectrometry(GC-MS)was used as analytical tools to extract volatile oil.The results: a The contents of Z-ligustilide,3-butylidenephthalide and n-butylphthalide of direct seeding Angelicae Sinensis is respectively22.12 mg/g,0.2791 mg/g and 0.0275 mg/g,and they are respectively 17.53 mg/g,0.2869 mg/g and 0.0248 mg/g in transplanting Angelicae Sinensis.Except 3-butylidenephthalide,the contents of other two ingredients in direct seeding Angelicae Sinensis are all higher than transplanting Angelicae Sinensis.b Except the contents of tyrosine,glycine,alanine and isoleucine is higher in direct seeding Angelicae Sinensis,the contents of other 11 kinds of amino acids were lower than them in transplanting Angelicae Sinensis.c The average contents of polysaccharides in direct seeding Angelicae Sinensis(0.466 mg/mL)is higher than it in transplanting Angelicae Sinensis(0.376 mg/mL).d There are 34 kinds of ingredients were extracted in volatile oil,the relative content of 3-butylidenephthalide in direct seeding Angelicae Sinensis(5.93%)is 1.62% higher than it in transplanting Angelicae Sinensis(4.31%),and the relative content of Z-ligustilide in direct seeding Angelicae Sinensis(72.38%)is 11.43%higher than it in transplanting Angelicae Sinensis(60.95%),but the relative contents of other ingredients(such as(1S)-2,6,6-Trimethylbicyclo[3.1.1] hept-2-ene,and so on),beta.-Pinene,.beta.-Ocimene,cis-.beta.-Farnesene,6-Undecanone is lower in direct seeding Angelicae Sinensis.Therefore,the contents of Z-ligustilide and 3-butylidenephthalide may be due to the difference quality of volatile oil in direct seeding and transplanting Angelicae Sinensis.C The comprehensive assessment of the quality of direct seeding and transplanting Angelicae SinensisTo comprehensive assessment of the quality of direct seeding and transplanting Angelicae Sinensis with TOPSIS.The result is that all the direct transplanting Angelicae Sinensis rank in the top five.And the quality difference of direct seeding and transplanting Angelicae Sinensis was analyzed by t-test,the results is that the contents of extract,volatile oils,ferulic acid and Z-ligustilide of direct seeding and transplanting Angelicae Sinensis have a extremely significant difference,the contents of total ash and acid-insoluble ash of them have a significant difference,but polysaccharide,3-butylidenephthalide and n-butylphthalide have no differences.The contents of other ingredients of direct seeding Angelicae Sinensis is better than that of transplanting Angelicae Sinensis except the contents of ferulic acid and3-butylidenephthalide.therefore,the main reason that lead to the low quality of seeding Angelicae Sinensis is the low content of ferulic acid and n-butylphthalide.D The comparative study on pharmacological effects of direct seeding and transplanting Angelicae SinensisTo compare the pharmacological effects of direct seeding and transplanting Angelicae Sinensis mainly by comparing CFU-GM and BFU-E cell colony formation,comparing the added value of spleen lymphocytes,comparing the ability of resistance DPPH free radical and hydroxyl free radicals.The results is that the promotion ability of direct seeding and transplanting Angelicae Sinensis decoction for the growth of bone marrow CFU-GM and BFU-E colony is almost the same,and there is no significant difference between both.After pleen lymphocytes were stimulated by ConA,the appreciation rate of pleen lymphocytes in direct seeding Angelicae Sinensis group(34.4%)is lower than it in transplanting Angelicae Sinensis group(35.7%),but there is no significant difference between both.The scavenging activity of seeding Angelicae Sinensis decoction on DPPH free radical is higher than transplanting Angelicae Sinensis decoction,but the scavenging activity of hydroxyl free radicals is lower than transplanting Angelicae Sinensis decoction.
