Objective: Bone defect repair had been a hot topic in the orthopaedic clinic,also been an important issue in the field of orthopedic research.Along with the research progress about the bone tissue engineering technology,the use of growth factors to promote healing of bone repair gradually become the research hot spot.Now a lot of literature reported platelet-rich plasma(PRP)had obvious bone induction ability and the ability to promote bone tissue repair,there were also some scholars were sceptical that the platelet-rich plasma had no significant ability to promote bone repair,this may be due to the metabolism of growth factors,The preparation of standard about PRP not unified and so on such causes.So it limited the platelet-rich plasma in orthopaedic clinical widely used,yet it's simple produced,rich source,and can provide necessary growth factor to bone repair,therefore the platelet rich plasma composite allogeneic bone graft to repair bone defect is a promising research direction.This experiment used autologous platelet rich plasma composite allograft to repair the defect in the distal femur,through the detection indicators to evaluate the ability of platelet-rich plasma to repair bone defect,in order to investigate the platelet-rich plasma composite allograft bone the curative effect for the treatment of bone defect,and provide guidance for clinical use.Methods:1 Each experimental rabbit was extracted about 5 ml of blood in the lithium heparin anticoagulant vacuum vessels on side of the ear vein,adopting secondary centrifugation,preparation of autologous PRP.2 A straight incision to be done at the rabbit's hind leg femur distal femoral condyle inside,to build a 7 mm diameter,depth of 10 mm and 5 mm from the knee joint line columnar defects by electric drill.3 A total of 36 New Zealand white rabbits were randomly divided into group A(12)and B(24).Group A: used for allogeneic bone preparation;Group B: divided into two subgroups,B1,B2.on the left side of the defect implanted PRP + pure cancellous allograft for B1 group,the right side of the defect implanted pure cancellous bone for B2 group.4 Postoperative observation generally,wound healing,etc.,at 4,8,12 w,group A randomly selected 4 rabbits,group B randomly selected 8 rabbits to be put to death,for gross observation,X-ray plain film,three color staining,HE staining and Masson shot density measurement,X-ray resistance and reference to Yang's radiological criteria to rate of each group of animal bone repair.5 Using Image analysis software(Image-Pro Plus6.0)to calculate the B1 and B2 group Masson trichromatic dye and HE staining red dye at 4,8,12 weeks differences by comparing two groups in the different time and using SPSS 13.0 statistical software for statistical analysis.To compare two groups the maturity degree and the activity of bone cells and new bone area of the proportion at different times.Results:1 Animals postoperative incision were healed well.2 X-ray beam density and radiological grading results showed that B1 and B2 group with statistical significance(P < 0.05)at 4,8 weeks,no statistical significance at 12 weeks(P > 0.05).3 postoperative 4,8 weeks,gross observation showed bone defect healing of B1 group is better than that of B2.4 HE staining(the percentage of red dye area of bone area)of group B1 was larger than group B2,with statistically significant at 4,8 weeks(P < 0.05),no statistical significance at 12 weeks(P > 0.05)5 Masson three trichrome stain of group B1 red dyed area was less than B2 group at 4 weeks,with statistical significance(P < 0.05),and 8 weeks after operation the dyed red area group B1 was greater than the group B2,with statistically significant(P < 0.05),postoperative 12 weeks,There was no different about group B1 and B2 at 12 weeks.Conclusions:1 PRP composite allogeneic bone can repair rabbit femoral defect in the distal lacunar.2 PRP composite allograft tissue could make the repair time shortened,cell activity high and the cells mature faster compared with pure implanted allograft.3 Little difference between the two groups in postoperative 12 weeks for lacunar bone repair.
