Part one Detection of differential proteins in saliva of patients with oral lichen planusObjective: Oral lichen planus(OLP)is a chronic oral mucosal disease of unknown etiology and long-term recurrent.The disease stubborn refractory and tendency to cancer.To investigate the differential protein expression in saliva of patients with OLP and healthy control group,and to analyze the expression of differential protein in saliva before and after treatment of different types of OLP.To explore the treatment effect of Huashi Xingyu Qingre decoction and the interaction mechanism of Huashi Xingyu Qingre decoction and OLP.Methods:1 Using ELISA to detect saliva of IgM,sAA,ZAG in OLP patients(20)and healthy subjects(20).2 ELISA was used for detecting the contents of IgM,sAA,ZAG in saliva of different type of OLP patients(10 erosion,10 non-erosion).3 The saliva of OLP patients(20)before and after Huashi Xingyu Qingre decoction treatment were measured by the methods of ELISA.4 The clinical curative effect of different types of patients with OLP(50erosion,50 non-erosion)on Huashi Xingyu Qingre decoction was observed.Results:1 The expression of IgM,sAA and ZAG in saliva of OLP group was higher than that in control group.The difference was statistically significant(P<0.05);2 Expression of two different types of oral saliva of patients IgM,sAA was not statistically significant.The expression of ZAG in saliva of oral lichenplanus erosive group was higher than that in textured group patients,and the difference was statistically significant(P <0.05);3 After therapy the expression level of IgM,sAA and ZAG in saliva was increased compared with before therapy,and the difference was statistically significant(P <0.05);4 This decoction for different types of OLP has a good therapeutic effect,the two groups were not statistically significant.Part two The expression of ABCG2 in the cancerous process of oral lichen planusObjective: In this study,we attempt to evaluate expression of ABCG2 in OLP tissues and OSCC tissues transformed from OLP,find effective molecular markers for predicting malignant transformation of OLP,and provide clues to find early treatment targets of malignant transformation of OLP.Methods:Immunohistochemical technology was used to detect ABCG2 protein expression in 30 tissue samples of OLP and 8 OSCC clinical tumor specimens transformed from OLP.We also compared and analyzed differences in expression levels of these proteins between OLP tissues and OSCC tissues transformed from OLP.Results:The ABCG2 positive staining was mainly expressed in the cytoplasm.Its expression in OLP tissue was weaker than that in OLP-OSCC tissue(Z=-4.103,P<0.05).Conclusions:1 Saliva in the IgM,sAA,ZAG may be involved in the pathogenesis of OLP.2 ZAG is highly expressed in the more erosive OLP stripe type,it may be involved in the development of OLP,and even deterioration process.3 The " Huashi Xingyu Qingre decoction " therapeutically saliva IgM,sAA,ZAG expression levels in patients before and after OLP significantdifference,suggesting that this prescription on the OLP has a good therapeutic effect,the mechanism needs further study.4 The clinical effect of treating different types of patients with OLP was significant.5 ABCG2 may be actively involved in the malignant transformation of OLP,ABCG2 high expression has a certain correlation with the malignant transformation of OLP.
