Objective: Thyroid cancer is the most common malignant tumor of the endocrine system.In the world,the incidence of thyroid cancer is significantly increased.So,to confirm the nature of thyroid nodules and early surgical treatment are the key points to achieve good prognosis.Recently searching for the diagnostic markers is a hot spot of current research.Some studies have shown that microRNA related to the occurrence of thyroid tumor development closely.But,the function of miR-221 and miR-222 is not clear in thyroid cancer.In this study,the expression levels of miR-221 and miR-222 in thyroid cancer tissues and the benign thyroid tissues were detected by quantitative real-time polymerase chain reaction,and analyzed the relationship between mi R-221 and mi R-222 and clinical pathological indexes.It would provide a new method for the diagnosis and treatment of thyroid cancer.Methods:1 The 65 cases of thyroid tissues were collected between July 2014 and July 2016 from the First Hospital of Hebei Medical University.All of the patients had not been accepted radiotherapy and endocrine therapy.There were 38 cases of benign thyroid tissues and 27 cases of thyroid cancer tissues.After the operation,the specimens were put in liquid nitrogen rapidly.2 The expression levels of miR-221 and miR-222 in thyroid cancer tissues and the benign thyroid tissues were detected by quantitative real-time polymerase chain reaction.3 The results of the experimental data were analysied by SPSS 19.0software.The data of non-normal distribution shows as P50(P25,P75),the comparison between two independent samples used non-parametric test.P <0.05 considered statistically significant.Results:1 The miR-221 expression had no significant difference between nodular goiter and thyroid adenoma [0.028(0.008,0.165)vs 0.053(0.009,0.170)](P> 0.05).The miR-222 expression had no significant difference between nodular goiter and thyroid adenoma [0.003(0.002,0.045)vs 0.008(0.002,0.041)](P>0.05).2 The expression level of miR-221 was higher in thyroid cancer tissues than in benign thyroid tissues[3.155(0.137,55.796)vs 0.030(0.007,0.162)](P<0.05).miR-222 expression was higher in thyroid cancer tissues than that in benign thyroid tissues[0.065(0.014,0.212)vs 0.006(0.002,0.038)](P<0.05).3 miR-221 and miR-222 expression had no significant correlation with the gender,age and classification(P > 0.05).miR-221 expression had no correlation with lymphatic metastasis(P > 0.05),while miR-222 expression had significant correlation with lymphatic metastasis(P<0.05).Conclusions:1 The expression levels of miR-221 and miR-222 was up-regulated in thyroid cancer tissues.2 The expression levels of mi R-221 and mi R-222 had no significant correlation between nodular goiter and thyroid adenoma.3 miR-222 participated in the regulation of the development of thyroid cancer.
