Objective The aim of this study was to research the effect of aqueous extract of Hippophae rhamnoides L.on the ability of antioxidant injury,proliferation,apoptosis and mitochondrial membrane potential of human cortical cells(Ha Ca T cells)by ultraviolet C.And to investigate whether the aqueous extract of Hippophae rhamnoides has protective function and its mechanism of UVC radiation in human immortalized keratinocytes.Methods The Ha Ca T cell lines were cultured in vitro.These cells were divided into these groups: the sham group(no irradiation,no drug),UVC model group(UVC radiation treatment,no drug),drug group(Respectively,adding 2mg /ml and 20mg/ml concentration of aqueous extract of seabuckthorn,5 minutes later giving UVC radiation).Then observe the morphological and quantitative changes of the cells in each experimental group.The activity of superoxide dismutase(SOD),glutathione peroxidase(GSH-PX),catalase(CAT)and malondialdehyde(MDA)content in Ha Ca T cells and supernatant was determined by enzyme-labeled method.The OD value of cell proliferation was determined by CCK-8 method.The apoptosis rate of each experimental group after Annexin V-PI staining and the mitochondrial membrane potential of JC-1 staining were detected by flow cytometer.Results Compared with the model group:with the increase in dosing concentration,the number of apoptotic cells decreased and the morphology became normal,The number of viable cells in each group was significantly different(P <0.05),and in a dose-dependent manner.With the increase in dosing concentration,the activity of SOD,GSH-PX,CAT were significantly increased,the content of MDA was significantly decreased.The consequents were positively correlated with the water extracts concentration of Hippophae rhamnoides.The difference between the groups was statistically significant(P <0.05).With the increase of dosing concentration,CCK-8 absorbance value increased significantly,and in a dose-dependent manner.The difference between the groups was statistically significant(P <0.05).With the increase of dosing concentration,the apoptosis rate decreased and the mitochondrial membrane potential increased.Conclusion The water extract of Hippophae rhamnoides has anti-oxidative damage to Ha Ca T cell line and inhibit Ha Ca T cell apoptosis,promote cell proliferation and increase mitochondrial membrane potential in a dose-dependent manner.Its mechanism may be related to its antioxidant capacity,protection of cell membrane structure and mitochondrial function.
