Purpose: The main factor affecting treatment and prognosis of clear cell renal cell cancer is metastasis. Now the metastasis mechanism of clear cell renal cell cancer is unclear. Lnc RNA TUG1(taurine upregulated gene 1) is a long non-coding RNA expressed in the retina and in the brain. The latest study shows that Lnc RNA TUG1 plays an important role in bladder cancer, gastric cancer, colorectal cancer, glioma,osteosarcoma, etc.This study mainly explore the expression of Lnc RNA TUG1 in cc RCC and paired normal tumor-adjacent tissues to investigate the correlations between Lnc RNA TUG1 and clinicopathologic characteristics for further study of Lnc RNA TUG1 in cc RCC.The study explore the affects of Lnc RNA TUG1 in cc RCC tumorigenesis and the molecule mechanism to support the theoretical basis of early diagnosis, accurate treatment and follow-up monitoring in cc RCC.Method:40 cc RCC specimens and paired normal tumor-adjacent tissues were random seleted from Shenzhen Second People’s Hospital’s specimen bank. Total RNA isolated from 40 cc RCC and paired normal tumor-adjacent tissues were used for obtaining c DNA by performing reverse transcription.Using RT-PCR to detect the Lnc RNA TUG1 expression level in renal cell carcinoma cell line A704,A498,786-0,ACHN and one normal human proximal tubule epithelial cell line HK-2. Picking out the two high expression renal cell carcinoma cell lines A704 and A498. Small interfering RNA(si RNA) was applied to suppress the Lnc RNA TUG1 expression in RCC cell lines(A489 and A704),then research the influence of slient Lnc RNA TUG1 to renal cell carcinoma cell lines A704 and A498’s biological behaviours such as growth, apoptosis,invasion and migration.Using CCK-8 to analyze cell growth.Using scratch assay to test cell migration ability, using Transwell experiment to test invasion ability to support new methods in early diagnosis, accurate treatment and follow-up monitoring in cc RCC.Results:(1)31 Lnc RNA TUG1 expresion of cc RCC were up- expression(77.5%).The Lnc RNA TUG1 expression in cc RCC were obvious high than in paired normal tumor-adjacent tissues(P<0.01);(2)There was none relationship between Lnc RNA TUG1 expression and age(P=0.457),gender(P=0.694),TNM(P=1.000),AJCC(P=0.453) and lymphatic metastasis(P=1.000).While it was related to tumor size and Forman grade;(3)The cell proliferation capacity of renal cell carcinoma cell lines A489 and A704 is reduce, cell invasive abilityis is decline sharply, cell apoptosis is bemarkedlyincreased while slienting Lnc RNA TUG1.Conclusion:(1)The high expression of Lnc RNA TUG1 in cc RCC may be related with tumorigenesis and progress. Clinical testing Lnc RNA TUG1 expression level may be good for the early diagnosis of cc RCC;(2) Lnc RNA TUG1 expression levels were associated with tumor size and Forman grade,it may used to predict tumor size and Forman grade;(3) Lnc RNA TUG1 may be the impartment oncogene in cc RCC tumorigenesis and progress. By inhibiting the expression of Lnc RNA TUG1 perhaps will become new accurate treatment of cc RCC.Purpose: To improve the clinicopathologic feature cognition of testicular malignant Leydig cell tumor(TMLCT) and level of non-invasive diagnosis. Methods :Retrospective analysis the clinicopathologic feature of TMLCT and detect the circulating tumor cell(CTC) in peripheral venous blood and review the related literature.Results : The patient underwent radical orchidoepididymectomy. Postoperation pathological diagnosis results revealled:testicular malignant Leydig cell tumor,tumor is mainly composed of leydig cells with suspicious vessel carcinoma embolus. It’s shown in immunohistochemical analysis that the tumor cells were positive for α-inhibin,Ki67,CD30,vimentin, EMA and PLAP, while the cells were negative for CK,CK7,S100,CD10,SMA,Des,AFP,HCG,CEA,CK19,CD117,Oct-4,LCA,CD20,Pax-5,CD3 and CD43.Two CTCs were detected in patient’s peripheral venous blood.Subsequently,the patient was received chemothearphy for three periods because of retroperitoneum multiple lymph nodes metastasis.The patient was underwent retroperitoneal lymphadenectomy and cryoablation by reason of the enlarged retroperitoneal lymph nodes in Computed Tomography.The patient ’ s retroperitoneal lymph nodes were obvious enlarged and right adrenal gland was invaded in 8 months follow up.Conclusion : TMLCT is extremely rare in sex gonad stromal tumor with high malignancy and poor prognosis,while CTC may be used for TMLCT’s early diagnosis and prognosis prediction.
