Objective To investigate the hypoglycemic effect of three kind crude separation portions from.To study and explore the chemical constituents of the extract of ginnala fruit and its hypoglycemic and antitumor activity,and to lay the foundation for the further development and utilization of tea maple resources.Methods Through the simple extraction and separation,five crude separation sites were obtained.Followed by the use of phenolic phenol and phenol-sulfuric acid method for the detection of various parts of ginnala fruit compounds content,and through the modern separation and analysis of various parts of the compound species and content determination.Hypoglycemic test ICR mice were fed with high fat diet and fasting streptozotocin(120 mg / kg),Mice with hypoglycemic levels of 11.1 mmol / L were randomly divided into control group,model group,acarbose group(25 mg/kg),activated charcoal boiling water elution high-and low-dose group,ethyl acetate high-and low-dose group,water extraction alcohol precipitation high-and low-dose group,10 rats in each group were given intragastric administration for 3 weeks.At the end of the experiment,Detection Indicator: blood sugar,blood lipids,serum insulin and the organ tissue was further observed by the staining.Anti-tumor experiment,H22 mouse hepatoma cells were inoculated with H22 mice in the left forelimb armpits to establish H22 tumor-bearing mice model,and successfully modeled mice were divided into,model group,cyclophosphamide group(30mg/kg,ip),activated charcoal boiling water elution high-medium and low-dose group,activated charcoal water elution high-medium and low-dose group,water extraction alcohol precipitation high-and low-dose group,ethyl acetate high-medium and low-dose group.Once a day,continuous administration of 10 d,which model group and blank control group were given the same amount of saline.At the end of the experiment,the tumor inhibition rate was calculated.The levels of serum IL-6,IFN-?,TNF-?,ALT,AST,BUN and CRE were measured by the kit to investigate the effects of ginnala fruit on the H22 transplantation tumor growth inhibition.The pathological changes of the tumor tissues were observed by he staining and the apoptotic cells were detected by TUNEL method,and to explore the anti-tumor mechanism of ginnala fruit by flow cytometry.Results The results showed that ginnala fruit was mainly composed of polyphenol,volatile oil and polysaccharide.Polyphenol mainly exists in the ethyl acetate site where the higher content of gallic acid,followed by quercetin;volatile oil mainly in the petroleum ether site,the higher content of the compound Nonacosane,followed by the compound Tetratriacontane.Polysaccharides mainly exist in the water to extract the sink site.THE three separate parts extrate from ginnala fruit can decrease the blood sugar levels of diabetic mice.Serum TC,TG,LDL-C,HbA1 c,MDA and BUN levels decreased significantly.Insulin,HDL-C,SOD and GSH levels were significantly increased.And there is a significant dose-dependent manner.The three crude separation sites were observed by staining.The liver and kidney tissues had a good protective effect.Among them ethyl acetate group is most significant.The three separate parts extrate from ginnala fruit can significantly inhibited tumor cell growth,among them activated charcoal boiling water elution group is most signific03.-ant.The four crude separation sites were observed by staining.The organ tissues had a good protective effect.And through the flow cytometry technology to find its anti-tumor mechanism is to induce tumor cell apoptosis and play a role in inhibiting tumor growth.At moment found that tea strips of fruit has improved immunity.
