Objective:This study analyzed the genotype and allele frequency distribution of CYP1B1 gene A119 S and L432 V in cervical cancer group and control group of Shanxi Datong area,and its correlation with the risk of cervical cancer.Furthermore,the relationship between polymorphisms and susceptibility to cervical cancer was analyzed by stratification of smoking(including passive smoking)and the differentiation and pathological types of cervical cancer cells.To find the genetic predisposing factors of cervical cancer,so as to better predict the occurrence and development of cervical cancer.Methods:This is a case control study.The 92 cases of cervical cancer patients who lived in Datong urban area and surrounding counties in Datong maternal and child hospital were selected as the case group,and 100 healthy women with normal TCT were served as the control group.The two groups were Han people and had no history of cancer.The age and smoking status of two samples,and the degree of differentiation and pathological types of cervical cancer patients were investigated.DNA was extracted from peripheral blood.We selected two site of polymorphism of CYP1B1 gene(119 site and 432 site),used PCR-restriction fragment length polymorphism(PCR-RFLP),enzyme digestion and gel electrophoresis.The data was analyzed through SPSS 17.0 software package,numeration data using ?2 test to check the difference in genotype distribution between patients and controls.The odd ration(OR)and 95% confidence interval(CI)represent the relative risk.Results:1.In Shanxi Datong Region,The genotype and allele frequencies of CYP1B1 A119 S and L432 V site were consistent with the Hardy-Weinberg principle(P>0.05)in the cervical cancer group and the control group,and were representative of the population.2.The frequencies of genotype GG,TG and TT in the CYP1B1 A119 S polymorphism were 55.4%,33.7% and 10.9% in Datong's individuals with cervical cancer,respectively.They were 66.0%,29.0% and 5.0% in controls individuals.there is no statistical difference between cervical cancer patients and controls(P>0.05).The frequencies of G and T in cervical cancer were 72.3% and 27.7%,respectively.The control group were 80.5% and 19.5%,there was on statistically significant difference between the two groups(P>0.05).3.The frequencies of CC,CG and GG in the CYP1B1 L432 V polymorphism were 50.0%,38% and 12.0% in Datong's individuals with cervical cancer,respectively.They were 70.0%,25.0% and 5.0% in controls individuals(P>0.05).there is statistical difference between cervical cancer patients and controls.homozygous mutant CC,heterozygous CG genotype compared with CC,the risk of cervical cancer was 3.35 folds(95%CI=1.09,10.27,P<0.05)and 2.13 folds(95%CI=1.13,4.02,P<0.05),respectively.The frequencies of allele C and G in cervical cancer were 69.0% and 31.0%,respectively.The control group were 82.5% and 17.5%,there was statistically significant difference between the two groups(P<0.05),Among them,the risk of cervical cancer with allele G is 2.12 folds of that of C(95%CI=1.03,3.42).4.In the smoking(including passive smoking),the genotype of CYP1B1L432 V containing G allele to cervical cancer risk increased by 1.31 folds(95%CI=1.08,4.95).there was statistical significance between cervical cancer group and control group(P<0.05);In nonsmokers,the genotype had no statistically significant difference between two groups(P>0.05).5.CYP1B1L432 V polymorphism in the differentiation degree of cervical cancer cells and different pathological types have no statistically significant difference compared with the control group(P>0.05).Conclusion:1.The polymorphism of CYP1B1 gene L432V(C-G)is related to the susceptibility of cervical cancer in Datong area of Shanxi,and may be independent of the polymorphism of A119S(G-T).2.In the CYP1B1 gene L432V(C-G)polymorphism,the genotype of the mutant allele G increases the risk of cervical cancer;and the higher the proportion of alleles G,the greater the risk of cervical cancer.3.Smoking(including passive smoking)and carrying a genotype of G at the L432 V site of the CYP1B1 gene increases the risk of cervical cancer.4.The polymorphism of CYP1B1 gene L432V(C-G)is independent of the degree of differentiation and pathological types of cervical cancer cells.
