Objective:Yixinnao Dripping Pills(YXNDP)were developed by our research group and had a role in protecting the myocardial and antihypertensive.In the study,the two-kidney one-clip(2K-1C)Goldblatt model of rats were used to investigate the antihypertensive effect and mechanism of YXNDP.Methods:The 2K-1C Goldblatt model of renovascular hypertension was used in SD rats.Renovascular hypertensive rats(RHR)were randomly divided into 5 groups: RHR group,captopril group(25 mg kg-1 per day orally),YXNDP-H group(2.0 g kg-1 per day orally),YXDNP-M group(1.0 g kg-1 per day orally),YXNDP-L group(0.5 g kg-1 per day orally),and sham group(untreated 2K-1C).8 rats in each group.Sistolic blood pressure(SBP)and diastolic blood pressure(DBP)were measured weekly when the rats were in awakened state.Followed up for 6 weeks,blood and tissue samples were collected for determination of nitric oxide(NO),total anti-oxidation capability(T-AOC),malondialdehyde(MDA),superoxide dismutase(SOD),catalase(CAT),glutathione peroxidase(GSH-Px).Plasma renin activity(RA),angiotensin I(AngI),angiotensin II(AngII)and endothelin(ET)levels were measured by enzyme linked immunosorbent assay.Results:Six weeks after 2K-1C operation,SBP of the model rats(163.0±6.6 mmHg)increased significantly compared with sham-operated rats(106.6±2.5 mmHg).SBP and DBP were significantly reduced by treatment with YXNDP.Meanwhile,the level of plasma ET was lower and NO was higher than those of control.YXNDP could increase the content of T-AOC and the activities of some endogenous antioxidative enzymes including SOD,CAT,GSH-Px in RHR.On the other hand,YXNDP could decrease the content of MDA,and suppresse intracellular ROS release in RHR.Additionally,plasma Ang II was markedly reduced after administration of YXNDP in 2K-1C hypertensive rats.Conclusion:These results provided substantial evidences that YXNDP had a good antihypertensive effect in 2K-1C hypertensive rats,which was likely related to through the anti-oxidate reaction,to improve the blood vessel endothelium function,and affected on rennin-angiotensin system activity.
