Objective:To investigate the effects of metformin on bone mineral density,biomechanics of the tibiae,and serum advanced glycation end products(AGEs)in ovariectomized(OVX)rats.Methods:Forty female SD rats underwent bilateral ovariectomy or sham operation,three months later they were randomly assigned into four groups for metformin treatment:Sham rats,OVX rats,OVX+50 mg/kg/day rats and OVX+100 mg/kg/day rats.Another three months later,all rats were sacrificed to evaluate the histology,dual energy X-rayabsorptiometry and biomechanics differences between different groups and to compare the serum AGEs differences by enzyme-linked immunosorbent assay(ELISA).Results:Compared with Sham group,bilateral ovariectomy induced impaired bone mass(P<0.05),dcreased bone strength(P<0.05),and increased serum AGEs(P<0.05);while metformin treatment significantly reversed these actions,leading to improved bone mass and decreased bone strength and serum AGEs(P<0.05).There was no significant difference between OVX+50 mg/kg/day rats and OVX+100 mg/kg/day rats in bone density and bone mass(P>0.05),however OVX+100 mg/kg/day rats showed increased bone strength when compared with OVX+50 mg/kg/day rats(P<0.05).Conclusion:1.Metformin may increase both bone mineral density and the biomechanics in ovariectomized rats.2.Metformin may improve bone metabolism by reducing AGEs in the serum of the postmenopausal osteoporosis rats. |
