Objecctive:To analysis the expression of p-Erk and Mps1 in B-RAFV600 E thyroid carcinomas and colorectal carcinomas,analysis its association with clinicopathological characteristics and prognosis.Methods:B-RAF mutation was detected by polymerase chain reaction-direct sequencing,statistical analysis the relationship between B-RAFV600 E and clinicopathological features and prognosis in thyroid carcinomas and colorectal carcinomas.We analyzed the presence of p-Erk and Mps1 in two malignant tumors by immunohistochemistry,analysis the relationship between B-RAFV600 E,p-Erk and Mps1.Result:1.The rate of B-RAFV600 E mutation was found in approximately 34.1% in thyroid carcinomas.It was significantly correlated with age,clinical T stage and N stage.The association of B-RAFV600 E mutation with OS(OS,Overall Survival)was not statistically significant.The expression of Mps1 was significantly increased in B-RAFV600 E mutations compared to that of B-RAFWT,and its expression was not correlated with p-Erk.2.The rate of B-RAFV600 E mutation was 5.2% in colorectal carcinomas.It was associated with differentiation and pathological type of colorectal carcinomas.The positive rate of p-Erk and Mps1 in colorectal carcinomas of B-RAFV600 E was significantly higher than that of B-RAFWT.The expression of p-Erk was significantly correlated with Mps1,and is closely related to differentiation,pathological type and lymph node metastasis.We found that B-RAFV600 E had lower overall survival than B-RAFWT patients by Kaplan-Meier survival analysis.Conclusion:Our findings suggest that Mps1 expression is associated with B-RAFV600 E mutation but not rely on the classic RAS/RAF/MEK/ERK signal pathway in thyroid carcinomas,it is possible to regulate the expression of Mps1 through unknown pathway.B-RAFV600 E is associated with low differentiated adenocarcinoma,especially associated with mucinous adenocarcinoma.As B-RAFV600 E patients had lower overall survival than B-RAFWT patients,B-RAFV600E/p-Erk can be used as an important reference to evaluate the clinical progress and prognosis of colorectal carcinomas.The expression of p-Erk was significantly correlated with Mps1 in colorectal carcinomas,suggesting that Mps1 may become a new potential therapy target for colorectal carcinomas.In order to elucidate the molecular mechanism of Mps1,this study provides a a new theoretical basis for molecular treatment of malignant carcinomas.
