| Esophageal carcinoma is one of common malignant tumors in human.It is highly occured in our country.The morality of esophageal carcinoma in our country also is the highest in the world.The changing of signal transduction exsists in mant kings of tumors.It is an important biological characteristic of tumor cells.In recent years,it has been indicated that the PI3K/Akt pathway plays an important role in the human cancers.What,more,we found than PI3 K and Akt protein are highly expressed in esophageal cancer and paly an imortant role in the growth and metastasis of esophageal caner.a number of research found the INPP4 B act as a tumor spuppressor in many tumors.Simlarly to PTEN,it act as tumor spuppressor through inhibition the PIK/AKT pathway.Objective To investigate the expression of inositol polyphosphate-4-phosphatase,type II(INPP4B),Akt and p Akt and explore its correlation to prognosis and clinicopathologic fators of esophageal cell carcinoma.Methods The expression of INPP4 B mRNA in 32 esophageal cancer tissues was detected by quantitative real-time PCR(q RT-PCR).The relationship between INPP4 B expression and clinicaland pathological characteristics was then analyzed Immunohistochemical analysis was performed to detect the expression of INPP4 B,Akt and p Akt proteins in 70 specimens of esophageal squamous carcinoma and 20 specimens of normal esophgeal tissues.The relationship of their expression with clinicopathological and prognostic parameters was analyzed.Result INPP4 B mRNA expression was higher in 32 esophageal cancer tissues than in adjacent normal tissues(P < 0.05).The expression of INPP4 B mRNA was not related to tumor differentiation,gender,age or tumor size、 lymph node metastasis and TNM stage in GC(P > 0.05).In 70 esophageal cancer tissues and 20 normal esophageal tissues,the rate of INPP4 B expression was significantly higher in esophageal cancer tissue than in norma esophageal tissue(67.1% vs 40%,P < 0.05),while the rates of Akt and pAkt expression were significantly higher in esophageal cancer tissue than in normal esophageal tissue(72.8% vs 25%;82.8% vs 15%,both P< 0.05).INPP4 B expression was nocorrelated with tumor differentiation,lymph node metastasis and TNM stage.Akt expression was correlated with tumor size,tumor differentiation,lymph node metastasis and TNM stage.PAkt expression was not correlated with phaogiocal factors in esophageal patients.INPP4B expressionwas positively correlated with Akt(r = 0.531,P= 0.000)and pAkt(r =0.327,P = 0.003)expression in esophageal squamous carcinoma.Patients with low INPP4 B expressionor or high Akt and pAkt expression had a significantly worse survival time than those with high INPP4 B expression or low Akt and p Akt expression(P = 0.000).Conclusion Both INPP4 B protein and mRN A are up-regulated in esophageal cancer,and its expression does not correlate with tumor differentiation,lymph node metastasis and TNM stage.Both Akt and p Akt protein are up-regulated in esophageal cancer,and its expression significantly correlates with tumor differentiation,lymph node metastasis and TNM stage.INPP4 B may be a tumor suppressor gene for esopheal cancer.The level of INPP4 B is positively correlated to progression and prognosis of esophageal cancer.The research may provide a new treatment for esophageal cancer.The expression of INPP4 B,Akt and p Akt proteins is closely related to the malignant biological behavior of HCC.INPP4 B can be used as a important factor for predicting prognosis in esophageal cancer patients. |
