Objective To investigate the expression of DC-SIGN and DC-SIGNR in cancer tissues of Hepatocelluar carcinoma(HCC),further more,to investigate the expression and clinical significance of sDC-SIGN in serum of patients with HCC.Method1.We detected the expression of DC-SIGN/DC-SIGNRmRNA in cancer tissues and paired adjacent tissues of 40 patients with HCC by real-time PCR.,and the protein expression of DC-SIGN/DC-SIGNR in 20 cases of HCC cancer tissue and paired adjacent tissues by immunohistochemical staining.2.We established the ELISA to detected the level of sDC-SIGN in serum of patients with HCC.We detected the alpha-fetoprotein(AFP)in serum of patients with HCC by electrochemiluminescence.We anlalysed the correlation between the sDC-SIGN level and the clinical pathological data of HCC patients by nonparametric statistical test(?2 test),and the consistency of distribution of sDC-SIGN and AFP in HCC by Kappa anlysis.We evaluated the diagnostic value of sDC-SIGN alone and combined with alpha fetoprotein(AFP)in the diagnosis of HCC by drawing ROC curve.Results1.The expression of DC-SIGN/DC-SIGNR m RNA and positive expression rate of protein in cancer tissues of HCC were significantly lower than those in adjacent tissues of HCC.2.There was not significantly correlation between the expression of DC-SIGN/DC-SIGNR mRNA in cancer tissues with age,sex,metastasis,invasion,tumor size,AFP and the positive of HBsAg(P>0.05).3.The level of sDC-SIGN in patients with HCC was lower than that in normal control group(P<0.01).4.When the rea under ROC curve(AUC)and cut-off value of sDC-SIGN were 0.673 and3.611ng/ml,respectively,the sensitivity and specificity for the diagnosis of HCC were66.8% and 60.9%,respectively.When the AUC of AFP was 0.845,the sensitivity andspecificity for the diagnosis of HCC were 66.7% and 87.8%,respectively.There was no consistency of distribution of sDC-SIGN and AFP in HCC by Kappa analysis(P>0.05).The combinated detection of sDC-SIGN and AFP could increase the AUC and sensitivity to 0.902 and 82.1%,respectively.5.There was no correlation of sDC-SINGR expression with age,sex,the positive of HBsAg,metastasis,and glisson capsule invasion of HCC patients(P>0.05).There was significant positive correlation between sDC-SIGN and tumor size(P<0.05).Conclusions1.The expression of DC-SIGN/DC-SIGNR was significantly lower in cancer tissues of HCC.It was suggested that DC function and T cell immune response were abnormal in HCC patients.2.The level of serum sDC-SIGN in patients with HCC was significantly lower than that in healthy population,and was positively correlated with tumor size,which could be used as a new serological marker for HCC diagnosis and disease evaluation.3.The combinated detection of sDC-SIGN and AFP could improve the diagnostic efficiency of HCC.
