ObjectiveStudy show that human genetic gene background has been related with infection of Human immunodeficiency virus, some special behavior, for example, drug abuse,unprotected sexual intercourse and STD affect the infection too. Our experiments study the gene diversity of the forth exon of DC-SIGN/DC-SIGNR in three groups(Normal people,people who have been infected by HIV-1 and ES) and the impact of special behavior on HIV-1 infection. We hope that we can analyzed HIV-1 infection from epidemiological factors and gene factors.MethodsWe collected three groups of sample: healthy people, people who have been affected by HIV-1, ES colony who were exposed in the condition of HIV-1, but blood test of HIV-1 antibody is seronegative. The content of the sample conclude the epidemiological data(sex, age, sex experience, experience of drug abuse, STD, etal) and blood sample. Genomic DNA is extracted from human blood by QIAamp DNA Blood Mini Kite Cat. We analyzed the gene diversity of the forth exon of DC-SIGN, DC-SIGNR by PCR and gelose gel electrophoresis. Sequence the variance genetype, then, we study the relationship between the gene diversity of DC-SIGN, DC-SIGNR and HIV-1 infection. We analyzed the effect of risk behavior on HIV-1 infection by chi-square test.ResultsWe have conceived a preparatory collection of illustrative plates of gene diversity (the forth exon repeated region ) of DC-SIGN/DC-SIGNR. We find a case of genotype 7/6 which has been reported that it can reduce the risk of HIV-1 infection for DC-SIGN, exists in Chinese people. Drug abuse, abusive sex and STD affect HIV-1 infection.ConclutionDC-SIGN/DC-SIGNR gene diversity existed in Chinese people, including the genotype 7/6 for DC-SIGN which has been reported that can reduce the risk of HIV-1 infection in Caucasian, while genotype 111 is the normal case in Chinese people, but it need more study to determine the function of the genotype 7/6 for yellow race. We also find that drug abuse, abusive sex, STD play an important role in HIV-1 infection.
