Calming The Liver And Relieving Pain Granule Effect Of On CGRP And 5-HTin A Rat Model Of Hyperactivity Of Liver Yang

Purpose: The analgesic and sedative effects of the flat liver pain relieving granule are verified by the related animal experiments and Mechanism of the action of calming the liver and relieving pain granule in the treatment of migraine.Study on the analgesic effect of particle on liver CGRP and 5-HT content of plasma in a rat model of hyperactivity of liver Yang.So as to provide theoretical basis and scientific basis of clinical application of the granule of calming the liver and relieving pain.Method: Hot plate experiment with Kunming mice,Acetic acid test and formalin induced pain test in order to test the analgesic effect of the granule.To observe the rate of the free activities of the mice in Kunming and the sleep rate of the mice in the cooperative hypnosis experiment,and to detect the sedative and hypnotic effects of the sedative granule.To observe the effect of the granule of calming the liver on the behavior of the model rats induced by nitroglycerin,and to determine the effect of the treatment of migraine.By detecting the changes of CGRP and 5-HT in plasma of rat model,the mechanism of the action of the granule of calming the liver is discussed.Result: There were no significant effect on pain threshold of mice induced by hot plate in four does group and positive medicine group.Compared with the control group,the high does,middle does group and the headache Ning capsule group could significantly reduced the number of twisting reaction induced by acetic acid in mice(p<0.05).There was no significant effect on the number of licking feet in the first phase of the mice in the high,middle,low does group and headache Ning capsule group.Headache Ning capsule and the low dose group can significantly reduce the number of licking feet in the second phase,compered with the control group(P<0.05orP<0.01).After used the high,medium and low dose group of the Calming the liver and relieving pain granule for 1,2,3and4 hours,there is no significant effect on the automatic activity of mice,and no significant difference(P>0.05)compared with the normal control group.Headache Ning capsule can reduce the activity of mice in the fourth hours compared with the normal control group,there is significant difference(P<0.05).The high,middle and low dose group and the headache Ning capsule group have the tendency to prolong the sleeping time of mice,and there is no significant difference between the control group and the control group(P>0.05).Flunarizine Hydrochloride Capsules can shorten the sleep time of mice,compared with control group(P<0.05).After subcutaneous injection of the Nitroglycerin,the model group CGRP content was significantly higher than normal control group,the comparison between the two groups has statistical significance(P<0.05orP<0.01).The levels of CGRP in high,medium and low does group were significantly decreased,and the difference between the model group and the model group was significant(P<0.001,P<0.01,P<0.05).The high,middle and low dose group of headache Ning and liver pain relieving granule could reduce the content of five serotonin,and there is a significant difference between the model group and the model group(P<0.05orP<0.01).Conclusion: Effects of safe and analgesic Granule on the content of CGRP and 5-HT in plasma of rats with migraine induced by subcutaneous injection of nitroglycerin.In this study,we showed that CGRP?5-HT content of plasma was significantly increased after the injection of Nitroglycerin.The levels of CGRP and 5-HT in plasma were significantly decreased in the high,medium and low dose group of Ping liver pain relieving granule.indicated that the Calming the liver and relieving pain granule can lower the activation level of CGRP and 5-HT in plasma of migraine model rats induced by nitroglycerin.CGRP has the function of expanding the cerebral blood vessels,while CGRP can produce inflammatory lesions of the blood vessels,the higher the content,the stronger the effect.but 5-HT only has vasoconstriction at high dosage,at the same time,it can inhibit the release of CGRP.however,5-HTD receptor activation in vascular expansion leads to headache,obviously decrease the content of 5-HT can alleviate the occurrence of migraine.Therefore,the Calming the liver and relieving pain granule inhibit abnormal cerebral vascular expansion through the affection on 5-HT and CGRP content.Restrain the exosmosis of plasma,platelet activation and other causes leading neurogenic inflammation,and to relieve migraine.Pinggan Zhitong granule has good effect of analgesia and sedation.Large dose of this medicine has a good effect of analgesia,and better effect of sedation.The study showed that liver pain particles can alleviate and treat migraine by analgesia and sedation.
