Lung cancer,as one of the tumors with the highest incidence and fatality rate,poses a severe threat to human health.So far,surgery,chemotherapy and radiotherapy are still the mostly applied counter measures for the clinical management of lung cancer.research.More and more research and development of targeted molecular inhibitor has made a certain progress of lung cancer treatment.However,patients with lung cancer are faced with poor prognosis with a 15%five-year survival rate.Precision medicirine is a prevailing trend leading current tumor treatment,the key to which is to identify effective biomarkers and distinguish target groups so that anti-tumor treatment can be customized for specific groups and gain higher efficiency.Therefore,it is essential to identify new effective molecular markers and potential treatable targets.Brand new understanding was obtained after the completion of Human Genome Project.It is safe to conclude that the lung adenocarcinoma(AD)and lung squamous carcinoma are two different diseases in the molecular and gene level.Growing evidences suggested that it may be more likely to find biomarkers in lung adenocarcinoma,and may be benefit from it.ASCL-1(Achaete-scute homolog-1)gene plays a critical role in development of the central nervous system,automatic nervous system,adrenal medullary chromaffin cells,thyroid C cells and pulmonary neuroendocrine cells.ASCL-1 gene expression is closely related to pulmonary neuroendocrine tumor,Studies have shown that excessive expression of ASCL-1 promote airway epithelial hyperplasia.The reports of relationship between ASCL-1 expression and prognosis were very few in lung AD.Objective Immunohistochemical and western blot was applied to detect the expressions of ASCL-1 protein in lung adenocarcinoma,explore the correlation between the expression of ASCL-1 and the clinicopathological parameters,the prognosis of AD patients.Methods ASCL-1 protein expression by immunohistochemical analysis in 283 cases of AD.Western blot was applied to detect the expressions of ASCL-1 protein in AD.Single factor chi square test and Logistic multiple factor regression analysis were used to explore the factors related to ASCL-1 expression.Kaplan-Meier survival analyses were used to evaluate prognosis of AD patients.Results The results of immunohistochemical analysis showed that ASCL-1 expression was positive in 48/283(16.9%)AD.Western blot test in 63 cases of AD and tumor-adjacent normal samples,9 cases of AD express ASCL-1 protein,and tumor-adjacent normal tissue without protein expression.By single factor chi square test and multivariate Logistic regression analysis,we found that the expression of ASCL-1 was associated with smoking(P=0.027),tumor differentiation(P=0.02),TNM staging(P=0.021)and lymph node metastasis(P=0.026)and overall survival(P<0.001).The overall survival time was shorter in the ASCL-1-positive group than in the ASCL-1-negative group by Kaplan-Meier survival analysis(P=0.02).Conclusion High expression of ASCL-1 protein is correlated with malignant phenotypes of AD,it is one of the independent prognostic factors for patients with AD...
