Objective: To investigate the expression of Nucleostemin(NS)in endometrioid adenocarcinoma,endometrioid adenocarcinoma paracancerous tissues and normal endometrial tissues,and to analyze the relationship between NS expression and clinicopathological features in endometrioid adenocarcinoma.Methods: Immunohistochemistry and Western blot were used to compare the expression of NS in endometrioid adenocarcinoma,endometrioid adenocarcinoma paracancerous tissues and normal endometrial tissues,and the relationship between the expression of NS and the pathological characteristics of endometrial adenocarcinoma was analyzed.Results:(1)Immunohistochemical results showed that in 90 cases of endometrioid adenocarcinoma,NS showed dark brown staining in endometrioid adenocarcinoma,and light brown staining or no staining in endometrial adenocarcinoma paracancerous tissues.NS protein was not expressed in 40 normal endometrial tissues;In endometrioid adenocarcinoma,the positive rate of NS expression(60.0%)was significantly higher than that in the paracancerous tissues(13.16%),and the difference was statistically significant(P < 0.001).In addition,The expression level of NS protein was significantly different in endometrioid adenocarcinoma tissues with different degrees of differentiation and FIGO stages,and whether there was lymph node metastasis(P < 0.05).Protein expression level of NS in endometrioid adenocarcinoma showed no statistical significance in the aspect of age,and whether menopausal(P >0.05).In endometrioid adenocarcinoma,age was not related to NS protein expression level(P >0.05).There was no relationship between whether menopause and NS protein expression level(P >0.05).(2)Western blot results showed that NS was not expressed in normal endometrium.The expression level of NS protein in endometrioid adenocarcinoma(3.091±1.104)was significantly higher than that in the paracancerous tissues of endometrial adenocarcinoma(0.693±0.467),and the difference was statistically significant(P < 0.001).In addition,the expression levels of NS protein were statistically significant in endometrioid adenocarcinoma tissues with different degrees of differentiation,different FIGO stages and whether there was lymph node metastasis(P <0.05).In endometrioid adenocarcinoma,age was not related to NS protein expression level(P >0.05).There was no relationship between whether menopause and NS protein expression level(P >0.05).Conclusion: NS was highly expressed in endometrioid adenocarcinoma,low in the paracancerous tissues of endometrioid adenocarcinoma,but not in normal endometrial carcinoma.The expression of NS protein may promote the development,metastasis and invasion of endometrioid adenocarcinoma,and may become a new target of molecular targeted therapy for endometrioid adenocarcinoma..
