Objective:To observe the effects of Yupingfeng granule on Th17/Treg imbalance and improving clinical symptoms in children with allergy to milk protein,and to explore the effect of Yupingfeng granule on Th17,Treg and its related factors in peripheral blood of children with allergic milk protein.Methods:Forty children with milk protein allergy were divided into two groups randomly:the conventional treatment group(20 persons),the conventional treatment+Yupingfeng granule group(20 persons).The health control group was composed of 10 normal children.The levels of IL-17,TGF-?1 were detected by the ELISA in the peripheral blood,the Th17 cells and CD4~+CD25~+Treg cells were measured by the flow cytometry,both on the normal children and on the milk allergic children.After diagnosis of milk protein allergy,the patients stoped to drink milk immediately,fed by breast milk or free amino acid milk powder;the mother who breastfeeds her baby should also avoid to drinking milk and any dairy products.The conventional treatment group was given oral probiotics and cetirizine syrup(6m-1y0.25mg/kg bid,1y-3y 2.5mg bid),,and rash patients additionally used the natri cream.The Yupingfeng granule treatment group was treated by the Yupingfeng particles,in addition to the conventional treatment(0.25mg/kg bid),for 2 months.The expression and proportion of Th17 cells and Treg cells in peripheral blood were measured by the flow cytometry.The levels of IL-17,TGF-?1 in serum were detected by enzyme-linked immunosorbent assay(ELISA).The ESAI score of the rash before and after treatment was recorded.SPSS 24 software was used to analyze the datum obtained before and after treatment.Results:The level of Th17 cells(2.1±0.7%,2.3±1.0%),the IL-17(34.63±7.52pg/ml?31.06±6.29pg/ml),in the conventional treatment group and the conventional treatment+Yupingfeng granule group were obviously increased in comparison with those of the normal children group(Th17 cells 1.1±0.5%,IL-17level 8.53±3.69pg/ml)while the corresponding Treg cells,and the TGF-?1 level were,respectively,(2.2±1.0%?2.4±0.7%)and(179.21±27.39pg/ml?181.18±30.03pg/ml),which are lower than those of the normal children(the Treg cells 3.5±1.0%,the TGF-?1 level353.11±41.19pg/ml),showing a statistically significant difference(P<0.05).There is no statistically significant difference between the conventional treatment group and the conventional treatment+Yupingfeng granule group in terms of the Th17 cells,the Treg cells,the IL-17 level,the TGF-?1 level and ESAI score(P>0.05).Regarding to the conventional treatment group,after treatment,the Th17 cells,the IL-17 level and ESAI score,were lowered to 1.6±0.6%,28.89±5.80 pg/ml,861.47±220.23pg/ml,and 4.61±2.60,showing a statistically significant difference(P<0.05),while the Treg cells and the TGF-?1 level were ascended to 2.9±0.8%and 224.40±45.23 pg/ml,showing a statistically significant difference(P<0.05).With respect to the conventional treatment+Yupingfeng granule group,after aretament,the Th17 cells,the IL-17 level,ESAI score,dropped to1.2±0.6%and 18.03±3.71 pg/ml,2.17±0.94,showing a statistically significant difference(P<0.05),while the corresponding the Treg cells and the TGF-?1level were increased to 3.5±0.9%and 340.07±34.96pg/ml,also indicating a statistically significant difference(P<0.05).Moreover,after the treatment,the Th17 cells and the IL-17 level,ESAI score,in the conventional treatment+Yupingfeng granule group were smaller than those of the conventional treatment group with a statistically significant difference(P<0.05);the Treg cells and the TGF-?1 level in the conventional treatment+Yupingfeng granule group were higher than those of the conventional treatment group with a statistically significant difference(P<0.05).Conclusions:(1).The Treg cells and its related factors,TGF-?1 are significantly decreased on milk protein allergic children,and the Th17 cells and its related factor IL-17 is significantly increased.(2).Probably,as an immunomodulator,the Yupingfeng granule can down-regulate the Th17 cells and IL-17,upregulate the Treg cells,TGF-?1,alleviate the allergic symptoms of milk protein.
