Purpose: To observe Y-box binding protein-1(YB-1)influence on m ultiplication,metastasis and invasion capacity of pancreatic cancer cel ls and the expression of YB-1,EMT-related molecules(E-cadherin,Vi mentin)in pancreatic cancer cell line PANC-1,and investigate prelim inarily the machanism of YB-1 affecting on invasion and metastasis of pancreatic cancer cells.Methods:We selected pancreatic cancer cells line PANC-1 for study.PANC-1 cells are transfected by YB-1 overexpression plasmid and plasmid HK for negative control by lipidosome-mediated way.There are three groups:blank group,blank load transfection group and YB-1 overexpression group.We detected the effect of overexpressed YB-1 on the multiplication,metastasis invasion capacity of pancreatic ca ncer cell line PANC-1 by MTT,Wound Healing and Transwell ex periment,respectively.The expression level of YB-1,E-cadherin,Vim entin and mRNA in pancreatic cancer cell line PANC-1 transfered b y YB-1 overexpression plasmid were determined using Western blot and RT-PCR.Results 1.MTT : The cell growth curve was determined by MTT method.T he result showed that the growth rate of YB-1 overexpression grou p was faster distinctly than blank group and blank load transfected group.It indicated that multiplication capacity of pancreatic cancer c ell line PANC-1 transfected by YB-1 over expression plasmid.(p<0.01);2.Wound Healing: The effect of YB-1 overexpression on panc reatic cancer cell line PANC-1' s multiplication capacity was detecte d by Wound Healing.Compared to blank group and blank load trans fection group,the mutiplication capacity of pancreatic cancer cell lin e PANC-1 transfected by YB-1 overexpression plasmid improved ob viously(p<0.01);3.Transwell invasion experiment: The result reveal ed that the invasion capacity affected on pancreatic cancer cell line PANC-1 of YB-1 over expression group was more remarkable tha n blank group and blank load transfection group.It indicated that th e invasive ability to pancreatic cancer cell line PANC-1 transfected by YB-1 overexpression plasmid increased apparently;4.Western blo t and RT-PCR: YB-1 and Vimentin' s expression level of pancreatic cancer cell line PANC-1 transfected by YB-1 overexpression plasm id were higher obviously than blank group and blank load transfecti on group,while E-cadherin 's expression was lower than blank grou p and blank load transfection group.(p<0.01)This illustrated indirect ly the up-regulated expression of Vimentin on pancreatic cancer cell line PANC-1 transfected by YB-1 overexpression plasmid,while the expression level of E-cadherin was down-regulated.Conclusion 1.Overexpressed YB-1 in pancreatic cancer cell line PANC-1 resulted in the improvement of multiplication,metastasis and invasion capacity of these cells.2.YB-1 might enable to down-regulate E-cadherin and up-regulate V-imentin by regulate EMT related molecules to facilitate the ability to multiplication,metastasis,invasion of pancreatic cancer cells.
