Expression And Clinical Significance Of YB-1 And EMT Related Proteins In Pancreatic Cancer

Object:To study the expression of the Y-box binding protein 1 and the Epithelial-mesenchymal transition related proteins E-cadherin and Vimentin in nonmalignant pancreatic tissue,pancreatic cancer tissue and adjacent tissue,and to analyze the three kinds of proteins relationship between the age,gender,tumor location,differentiation degree,TNM stage and lymph node metastasis of pancreatic cancer.Method: Respectively using immunohistochemical method and Western blot to detect the expression of the YB-1 and the E-cadherin and the Vimentin in 30 cases of surgical resection of pancreatic cancer tissue and its corresponding tissue adjacent to carcinoma and 10 cases of surgical removal of the same period in nonmalignant pancreatic tissue.Result:1.Immunohistochemical results show:(1)The positive rate of YB-1expression in pancreatic cancer tissue was 70%(21/30),in tissue adjacent to pancreatic cancer was 30%(9/30),while in nonmalignant pancreatic tissue was 0%(0/10).There was a difference between pancreatic cancer group and nonmalignant pancreatic group and By the cancer group(all P<0.01).While there was no significant difference between nonmalignant pancreatic group and By the cancer group(P>0.05).(2)The positive rate of E-cadherin expression in pancreatic cancer tissue was 43.33%(13/30),in tissue adjacent to pancreatic cancer was 90%(27/30),while in nonmalignant pancreatic tissue was 100%(10/10).There was a difference between pancreatic cancer group and nonmalignant pancreatic group and By the cancer group(all P<0.01).While there was no significant difference between nonmalignant pancreatic group and By the cancer group(P >0.05).(3)The positive rate of Vimentin expression in pancreatic cancer tissue was 56.67%(17/30),in tissue adjacent to pancreatic cancer was23.33%(7/30),while in nonmalignant pancreatic tissue was0%(0/10).There was a difference between pancreatic cancer group and nonmalignant pancreatic group and By the cancer group(all P<0.01).While there was no significant difference between nonmalignant pancreatic group and By the cancer group(P>0.05).(4)The expression of the YB-1 and the E-cadherin and the Vimentin were highly correlated with differentiation degree,TNM stage and lymph node metastasis(all P<0.05),while uncorrelated with patients' gender,age and tumor location.2.Western-blot results show:(1)The protein expression level of YB-1 was significantly increased in pancreatic cancer group compared with the nonmalignant pancreatic group and By the cancer group(all P<0.01).While there was no significant difference between the last two groups(P>0.05).(2)The protein expression level of E-cadherin was significantly decreased in pancreatic cancer group compared with the nonmalignant pancreatic group and By the cancer group(all P<0.01).While there was no significant difference between the last two groups(P>0.05).(3)The protein expression level of Vimentin was significantly increased in pancreatic cancer group compared with the nonmalignant pancreatic group and By the cancer group(all P<0.01).While there was no significant difference between the last two groups(P>0.05).Conclusion:(1)YB-1 and Vimentin were highly expressed in pancreatic cancer tissue,while E-cadherin was low in pancreatic cancer tissue.(2)The high expression of YB-1 and Vimentin in pancreatic cancer tissue and the low expression of E-cadherin in pancreatic cancer tissue were associated with histological differentiation,TNM stage and lymph node metastasis in pancreatic cancer,suggesting that the three may be involved in the occurrence,development,invasion and transfer of pancreatic cancer.(3)The incidence expression of the YB-1 and the E-cadherin and the Vimentin are closely related with the development of pancreatic cancer,the expressed of YB-1 in pancreatic cancer may participate in the regulation of EMT make the expression of E-cadherin cut and Vimentin expression raised to promote the development and metastasis of pancreatic cancer.
