purpose : The micelles of indomethacin / FA-HA-VE polymer were prepared by targeting the CD44 receptors of folate receptors,and the drug-loaded micelles were characterized.And to study its in vitro release and rat pharmacodynamics.Methods : 1.Preparation of FA-HA-VE: introduction of amino groups with tocopherol succinate as raw material and ethylenediamine to form amide bond,activation of hyaluronic acid carboxyl group with EDC,NHS,synthesis of HA-VE with amino vitamin E,activation with EDC.2.Preparation of FA-HA-VE micelles containing IMC : IMC / FA-HA-VE micelles were prepared by solvent evaporation and dialysis.The results showed that solvent evaporation method was higher than that of dialysis method.The ratio of different drugs to carrier was optimized.When the drug reached a certain proportion,Drug loading is no longer increased.The drug loading and encapsulation efficiency of HA-VE were 40.25 ± 4.36% and 10.73 ± 1.12% respectively.The drug loading and encapsulation efficiency of FA-HA-VE were 34.36 ± 2.56% and 9.16 ± 0.94% respectively.The particle size of the two is 126.9 ± 5.0 nm 135.0 ± 8.5nm,the potential is-22.4 ± 1.4Mv,-15.8 ± 1.0mv,and the micelle size is narrow.3.Study on the release of drug-loaded micelles in vitro by dialysis :Mainly compare the cumulative release rate of HA-VE and FA-HA-VE in vitro.Compared with free drug,HA-VE and FA-HA-VE containing IMC had sustained release effect,but there was no significant difference between them.
