Background:Colorectal cancer is one of the malignant tumors with high morbidity and mortality in China.Currently,the treatment of colorectal cancer is based on surgical resection,supplemented by chemotherapy,radiotherapy,immunotherapy,traditional Chinese medicine treatment,targeted therapy,aiming to improve the therapeutic effect.Over the years,with the continuous in-depth study on targeted therapy,some progress has been made in the.targeted treatment of colorectal cancer,but the treatment is still not successful.Finding genes that promote or inhibit tumor growth is the basis for the achievement of targeted therapies.Protein deglycase DJ-1,which was found in autosomal recessive early-onset familial Parkinson s disease,so it was also known as Parkinson s disease–related protein-7.It was frst identifed as an oncogene in cooperation with H-RAS and caused malignant transformation of mouse NIH3T3 cells Moreover,DJ-1 has been found in various cancerous tissues,including medulloblastoma,glioma,lung cancer[7],and oral squamous carcinoma.Abundant DJ-1 was shown to promote the growth and multidrug resistance of tumor cells by downregulating the expression of bax and inhibiting caspase activation.On the other hand,DJ-1 was reported to inhibit melanoma metastasis into the lung by decreasing the levels of growth hormone,suggesting that the role of DJ-1 in tumor progression is controversial.However,there is no information concerning the functional and clinical value of DJ-1 in CRC.Objective:Therefore,in this study,the expression of DJ-1 protein in colorectal cancer tissues was studied by immunohistochemistry,and adjacent tissues and normal colorectal tissues were established as controls.The differences in the positive rate and expression level between the groups were compared.The relationship between DJ-1and the occurrence and development of colorectal cancer was explored in order to analyze its relationship with clinicopathological parameters of colorectal cancer,aiming to provide a new approach for targeted treatment of colorectal cancer and provide a new line for judging the prognosis of colorectal cancer.Methods:Tissue slice and immunohistochemistry?SP method?were used to Detect the expression of DJ-1 in normal tissue,paracancer tissue and colorectal carcinoma tissue.Using the?2 test and the Fisher test and analysising the data with Spss software,the significance was set at P<0.05.Results:The expression positive rate of DJ-1 in normal tissue,adjacent mucosa and colorectal cancer was 74/99?74.74%?,8/99?8.08%?,1/15?6.67%?respectively,the difference between the groups has statistical significance?P<0.05?.The expression in colorectal cancer tissues was significantly higher than that in adjacent tissues?P<0.05?.The expression positive rate of DJ-1was12/18?66.67%?,7/8?87.50%?,9/14?64.29%?,19/25?76.00%?,26/34?76.47%?,respectively in Horizontal Colon cancer,Ascending colon carcinoma,Descending colon cancer,Sigmoid colon cancer,Rectal carcinoma.The expression positive rate of DJ-1was 39/51?76.47%?and35/48?72.92%?respectively,in tumor diameter greater than 5 cm and tumor diameter less than 5cm.The expression positive rate of DJ-1was16/18?88.89%??17/19?89.47%??42/62?67.74%?,respectively,in well-differentiated adenocarcinoma,moderately differentiated adenocarcinoma and poorly differentiated adenocarcinoma,difference between the groups has no statistical significanc?P<0.05?.The expression positive rate of lymph node metastasis group was 33/36?91.67%?significantly higher than the group without metastasis41/63?65.08%?,the difference between the groups has statistical significance?P<0.05?.And comparing stage N1and N2,the expression positive rate of DJ-1was 29/30?96.67%?and2/5?40.00%?respectively,the difference between the groups has statistical significance?P<0.05?.The expression positive rate?+?stage of TNM was33/36?91.67%?significantly higher than of?+?stage41/63?65.08%??P<0.05?.The expression of DJ-1 has no significantly difference between men and women.?P>0.05?.According to results of data analysis Spearman rank correlation,it shows that there is significant differences in the positive or negative expression of DJ-1 between normal colorectal tissue,adjacent mucosa and colorectal cancer tissue.Conclusion:The overexpression of DJ-1 related to lymph node metastasis and clinical stage in colorectal cancer.,It was no related to tumor tissue differentiation,tumor size,tumor location and patient gender age.It has certain reference value to further study the molecular mechanism of the occurrence and development of colorectal cancer.It may also serve as a new diagnostic marker and therapeutic target for colorectal cancer.
