Objective:To evaluate the effects of non-thermal plasma combined with X-ray on A549 and Hela cells.Methods:Cells were divided into four groups,control group(Control),non-thermal plasma treatment group(NTP),irradiation group(IR)and plasma plus irradiation group(NTP+IR).After treatment of plasma and irradiation,cell migration ability was detected by the scratch assay,cell invasion ability was detected by transwell assay.Protein expression was detected by Western blot assay.Results:1、The results of wound healing assay for detection of migration ability of A549 cells and Hela cells:A549 cells:The rate of migration of Control group 100.0%、NTP group(52.6±4.3)%、IR group(54.6±3.0)%、NTP+IR Group(10.8±2,8)%。Hela cells:The rate of migration of Control group 100.0%、NTP group(62.5±3.6)%、IR group(61.6±1.4)%、NTP+IR group(31.0±2.9)%.2、The results of Matrigel invasion assay for detection of invasion ability of A549 cells and Hela cells:A549 cells:The rate of invasion of Control group 100.0%NTP group(52.4±11.0)%、IR group(79.1±7.8)%、NTP+IR group(38.4±3.6)%.hela cells:The rate of invasion of Control group 100.0%、NTP group(28.6±3.2)%、IR group(26.7±7.3)%、NTP+IR group(15.0±6.7)%.3、The results of Western Blot for detection of the protein levels of p-paxillin、p-FAK and MMP-9:Either X-ray or NTP can down-regulate the expression of MMP-9 and the phosphorylation of Paxillin and FAK.The degree of down-reglation of combined group is stronger than single X-ray group.Conclusion:1、NTP can enhance irradiation-induced suppression of migration and invasion of A549 cells and Hela cells.2、The suppression could be associated with the down-regulation of the expression of MMP-9 and the phosphorylation of Paxillin and FAK.
