Objective: The purpose of this experiment is to research the relationship between BRAF V600 E and RAS mutation and PTC and to discuss the difference correlation among the non-transfer papillary thyroid carcinoma group,the papillary1.thyroid carcinoma group with regional lymph nodes metastasis and the papillary thyroid carcinoma group with distant metastasis.Methods: The tumor tissue specimens of 86 patients with highly differentiated PTC are collected from the Hunan Cancer Hospital,and the amplification refractory mutation system(ARMS)method is used to detect BRAF and RAS gene(NRAS and KRAS).SPSS22.0 software is used for data analysis.The chi-square test(?2 test)is performed on 86 case data single factors.Secondly we use Binary Logistic multivariate analysis to analyze the factors mentioned above.Results: 1.It is obtained that the mutation rate of BRAF is 54.65%(47/86),all of which are V600 E mutation.The mutation rate of NRAS is 5.81%(5/86),One case has No.3 hole mutation(G13R,G12 C,G12V,G12 A and G13V),three have the 61 codonmutation of RAS gene(Q61R,Q61 K,Q61Land Q61H),and another has A59Dmutation;whereas the mutation rate of KRAS is 0.00%.In non-transfer well-differentiated papillary thyroid carcinoma group,the BRAFV600 Emutation rate is57.69%(15/26);well-differentiated papillary thyroid carcinoma group with regional lymph nodes metastasis with the BRAFV600 Emutation rate of 70.00%(21/30);and well-differentiated papillary thyroid carcinoma group with distant metastasis with the BRAFV600 Emutation rate of 36.67%(11/30).2.In the single factor analysis,it is obtained that BRAF V600 Emutation is not statistically significant with patient's gender and tumor size(taking 2cm as the standard),and whether there is a lymphatic metastasis(p> 0.05),however,it is statistically significant with age(p=0.017),whether there is distant metastasis(p=0.014),staging(p=0.027)and whether it is minimum cancer(<1 cm)(p= 0.026).There was significant difference in the mutation rate among the non-metastasis group,the local lymph node metastasis group and the distant metastasis group(p=0.032).3.In the multivariate analysis,we find out that BRAF V600 E gene mutation has some relationships with distant metastasis(p=0.001)?pathological staging(???/???)(p=0.011)andmicro-carcinoma(p=0.044).Conclusions:1.In86 well differentiated PTC patients,BRAF gene'smutation rate is higher than RAS gene's in well differentiated PTC patients.2.There is a independent correlation between BRAF V600 E gene mutation and distant metastasis?pathological stage and micro-carcinoma.(p<0.05).3.The mutation rate of the local lymph node metastasis group is higher than the non-metastasis group,and the distant metastasis group.
