The Influence Of Sodium Ferulate On Expression Of TGF-?1,?-SMA In CCL4 Induced Rat With Liver Fibrosis

Objective:In this article,we built the liver fibrosis rat model by intraperitoneal injection with carbon tetrachloride?CCl4?to explore the expression of proteins TGF-?1,?-SMA on theliver tissue of SD rat with liver fibrosis,as well as the effect of sodium ferulate on that expression.With the initial investigation on mechan-ism of actionof anti-fibrosis by sodium ferulate,theoretical basis will be pro-vided for clinical therapy of sodium ferulate on liver fibrosis.Methods:In this experiment,we selected 48SD rats,among which 10 were ran-domly chosen into the blank control group,and the rest of38rats were given CCl4 for constructing the liver fibrosis modelby fasting intraperitoneal injec-tion.After four weeks,2 rats were killed at each two weekends to observe pa-thological changes by Hematoxylin and Eosin staining.Hepatic fibrosis in rats emerged at the end of the eighth week.The survived 26 rats could be di-vided equally into Fibrotic model group and Sodiumferulate treat group.Treatment group adopted sodiumferulate by instragastric administration?so-dium ferulategiven as 100mg/kg?,while fibrotic group and blank control group were given 0.9%NS at the same time.All the rats were given 40%CCL4and olive oil until the end of the experiment except for control group.At the end of 12^th week,all the rats were executed.ALT?AST?ALB and TBIL were detected by automatic biochemistry equipment,and liver fibrosis was eva-luated by HE staining and Masson staining.The expression of TGF-?1 and?-SMA was detected by Western boltting and immunohistochemistry?IHC?methods.Results:1.The serum level of ALT?AST?TBIL,the pathological fibrosis scores,as well as the expression of TGF-?1 and?-SMA in live of rats in fibrotic mod-el group induced by CCL4 all increased significantly compared with normal group;however,the serum levels of ALB decreased significantly compare with normal group.2.The serum levels of ALT?AST?TBIL,the pathological fibrosis scores,as well as the expression of TGF-?1 and?-SMA in live of rats in treatment group all increased significantly compared with Fibrotic model group;howev-er,the serum levels of ALB decreased significantly compare with Fibrotic model group..Conclusions:1?Sodium ferulate can reduce the expression ofTGF-?1 and?-SMA in the rats with liver fibrosis.2?Sodium ferulate improve liver fibrosis probably by decreasing the expression level of TGF-?1 and?-SMA.
