| Chronic damage to the liver by variety of causes frequently results in fibrogenesis,the increased deposition of extracellular matrix(ECM).The main component of the newly formed ECM is collagen.Hepatic stellate cells,when transformed into myofibroblasts,are the major source of this newly synthesized collagen.In the long run,the pathological accumulation of collage disrupts the organ抯 lobular structure and impairs hepatic function,frequently accompanied by incresed portal vein pressure and reduced hepatic blood flow.The end-stage of fibrogenesis is cirrhosis, a disorder for which no specific treatment currently. Recently ,it is known to all that free radical plays a important role in the process of liver injury and hepatic fibrosis. Free radical can activate HSC, facilitate ECM accumulation . In this study ,we investigate the anti-peroxdation effects of Sodium Ferulate (SF) on acute liver injury in mice and its anti-fibrosis in rats induced by carbon tetrachloride(CC14). Acute liver injury in mice was induced by CC14 and D-galactosamine(D-Ga1N),and treated with SF. Histopathologic change in liver tissue,serum alanine aminotransferase(ALT) , asparate aminotransferase (AST) and Malondialdehyde (MDA), Superoxide Dismutase (SOD), glutathione peroxidase (GSH-Px) in the liver were observed. The results revealed that elevated serum ALT,AST activities and MDA contents in liver tissue were markedly decreased, and the decreased SOD, GSH-Px activities in the liver were improved with SF treatment. Histopathologic change in liver tissue were alleviated in mice with administration of SF.The actions of SF on acute liver injury were similar to those of biphenyldicarboxylate (PBD). In the further study,we wanted to investigate whether SF can prevent and delay the development of hepatic fibrosis in rats.Male Sprague-Dawley(SD) rats were used.Hepatic fibrosis model was induced by injecting Cd4 ,dissolved in olive oil 1 :1(vol/vol),lmlIkg body weight, subcutaneously once every three days . Rats in SF prevention group were given SF at first injection of CCI4. Rats in SF treatment group were given SF after six weeks of injecting CC14.The doses of Cd4 and SF was adjusted weekly based on changes in body weight.All rats were killed at the thirteenth week. Serum ALT ,AST, bilirubin(BIL),hyaluronic acid (HA), Laminin (LN) were tested. SOD activities, MDA and Hyp contents in the liver tissue were observed. Specimens were taken and prepared for light and electron microscopic examination. Histopathologic change in liver tissue were observed with HE stain, Masson stain, Sirus red stain and reticular fiber stain respectively. The extra-structure of the rat'S liver were observed with electronic microscope. The results showed that SF ameliorated CC14-induced hepatic fibrosis significantly. The liver Hype MDA contents and Serum ALT, AST,BIL, HA,LN were reduced in the rats treated with SF, on the other hand, those in the hepatic fibrosis model group were obviously higher. The liver SOD contents in SF group was higher than that in model group. The histological examination also demonstrated that SF had anti-fibrotic effect. The liver in most of rats in pathologic model group formed pseudolobule ,the pathology change of collagenic fibers and reticular fibers in liver with special histological stain were alleviated in SF group.The collagen of liver tissue were higher in model group than SF group observed with optic microscope.
