Obj ective:Observe the curative effect of KeLiu Pill in treatment of colorectal cancer,the quality of life,and the side effects of chemotherapy;analyze the effect of Keliu Pill on the short-term curative effect,quality of life,and side effects of chemotherapy;observe the Anti-tumor effect of Keliu Pill in PDTX model.Preliminary attempt to use PDTX model as the research object to explore the anti-tumor effect of KeLiu Pill will provide ideas for subsequent researchers to study Chinese medicine by using PDTX model.Methods:1.Forty patients with advanced colorectal cancer diagnosed by cytology or pathology were randomly divided into treatment group(KeLiu Pill combined with FOLFIRI)in 20 cases and control group(FOLFIRI)in 20 cases.After 4 courses of treatment,the two groups were observed.The recent objective changes in effectiveness,colorectal cancer-related tumor signs,TCM symptoms and KPS scores,and the incidence of toxic and side reactions during chemotherapy in both groups are observed.2.The PDTX model of colorectal cancer was established in NCG mice and divided into normal saline group(negative control group),irinotecan group(positive control group),and KeLiu Pill group.The daily activities and spirits of the mice during administration and changes in status and tumor growth,were observed.Calculation of tumor volume,tumor weight inhibition rate.Results:1.Clinical efficacy:RR comparison,P=0.705>0.05,DCR comparison,P=0.429>0.05,the two groups of data were statistically processed,and the differences were not statistically significant.There was no significant difference in the two groups of tumor markers before and after treatment.For the improvement of TCM symptoms,the total score of the treatment group was higher than that of the control group,P=0.008<0.05,the difference was statistically significant;the total effective rate of the improvement of the syndrome in the treatment group was higher than that of the control group,P=0.025<0.05,the difference was statistically significant significance.In terms of physical status,the total improvement rate of KPS score in the treatment group was higher than that in the control group,P=0.038<0.05,and the difference was statistically significant(P<0.05).Regarding adverse reactions,the total incidence of gastrointestinal adverse reactions such as myelosuppression and diarrhea in the treatment group was lower than that in the control group,with a significant difference(P<0.05).2.Experimental part:The tumors in the Keliu Pill group were significantly larger than the saline group and greater than the irinotecan group.The difference was not statistically significant(P<0.05);the tumor volume of the Keliu Pill group was greater than that of the normal saline group and the irinotecan group.There was not statistically significant difference(P>0.05).The quality of life of mice KeLiu Pill group was better than other two groups.Conclusion:KeLiu Pill has certain curative effect in controlling tumor progression and improving the quality of life of patients,and has no obvious side effects.Combined with chemotherapeutic drugs can improve the side effects after chemotherapy to some extent.PDTX model can completely maintain the original pathophysiological characteristics of patients with primary tumors,and can remain stable in various passages.Keliu Pill had no obvious effect on inhibiting the growth of subcutaneous transplanted tumors in colorectal cancer model,but compared with irinotecan,there was no significant side effect on the quality of life of mice.
