Eucommiae Cortex is a unique and valuable medicinal material in our country.It comes from the dried bark of Eucommia ulmoides Oilv.It has the effect of supplementing liver and kidney,strengthening muscles and bones,and protecting the fetus,so it can be used for deficiency of liver and kidney,pain of waist and knee,weakness of muscles and bones,dizziness,bleeding from pregnancy,restlessness of fetus,and so on.However,factors such as origin,collection,processing,'sweating' method and other factors have a certain impact on the effective ingredients of Eucommia ulmoides.How to ensure the quality of Eucommia ulmoides has become a key problem in the production process.Through the systematic analysis of the multi-functional constituents in Eucommia ulmoides,we compared the samples from different habitats,from different harvest times,processed by different methods and different ' sweating'conditions and different medicinal parts of Eucommia ulmoides.UFLC-Triple TOF-MS/MS technique has been used to analyse chemical constituents and their differences of the samples and find out the change of their quantity.Fluorescence in situ Hybridization(FISH)technique was used to analyse the changes of microbial community structure in samples of different stages of 'sweating'.It provided basic data and scientific basis to explore the formation process of Eucommia ulmoides medicinal materials quality,chose suitable production and breeding,cultivation and appropriate reasonable harvesting and processing.The research contents include the following aspects:1.Literature research The literature research system consulted the relevant literature of Eucommia ulmoides at home and abroad,and carried on the literature review research to it.2.The chemical constituents of Eucommiae Cortex were analyzed by UFLC-Triple TOF MS/MS.According to the accurate molecular weight and the fragment ion peak obtained by high resolution mass spectrometry.The retention time of chromatography and the information of reference substance were used to identify the 35 constituents,and the possible cleavage pathways and rules were deduced.To further explore the efficacy of Eucommiae Cortex medicinal material basis,quality changes and quality control mechanism.3.Quality analysis of Eucommiae Cortex from different areas and commodity.UFLC-QTRAP-MS/MS was developed for the simultaneous determination of 14 constituents in Eucommiae Cortex from different origin and commodity.ICP-OES was used for the simultaneous determination of 24 kinds of inorganic elements in Eucommiae Cortex from different habitats.UFLC-Triple-TOF-MS/MS was used to analyse the chemical constituents and the differences of Eucommiae Cortex from different habitats.The results showed that there were significant differences in 14 constituents between Eucommiae Cortex from different habitats and commercial medicinal materials.There was a significant difference in inorganic elements contents between Eucommiae Cortex from different habitats.There were obvious differences in the chemical compositions of Eucommiae Cortex from different habitats,and 14 kinds of common chemical constituents were identified through the analysis.4.Study on the changes of the dynamic accumulation of secondary metabolites of Eucommiae Cortex.UFLC-QTRAP-MS/MS was used for the simultaneous determination of 14 constituents in Eucommiae Cortex from different harvest time.ICP-OES was used for the simultaneous determination of 24 kinds of inorganic elements in Eucommiae Cortex from different harvest time.UFLC-Triple-TOF-MS/MS was used to analyse the chemical constituents and the differences of Eucommiae Cortex from different harvest time.The results showed that there were obvious differences among 14 index constituents of Eucommiae Cortex at different ages and harvest times.Grey correlation analysis showed that the better age of Eucommiae Cortex is 7?10 years old,and the quality of Eucommiae Cortex is better in early June.Inorganic elements showed dynamic changes in Eucommiae Cortex at different ages and different harvest times,and the chemical composition of Eucommiae Cortex in different ages and different harvest times were obviously different.The chemical difference of Eucommiae Cortex was analyzed by PLS-DA.18 and 12 different chemical constituents were identified respectively.5.Study on the effects of different processing methods and 'sweating' conditions to Eucommiae Cortex.UFLC-QTRAP-MS/MS was used for the simultaneous determination of 14 constituents in Eucommiae Cortex processed by different methods and different 'sweating'condition.UFLC-Triple-TOF-MS/MS was used to analyse the chemical constituents and the differences of Eucommiae Cortex processed by different methods and different 'sweating'condition.The results showed that the contents of 14 index constituents in Eucommiae Cortex under different processing methods and 'sweating' conditions were obviously different,and the grey correlation analysis showed that the quality of 'sweating' and "Directly stacking sweating with scraping off rough skin" was better.Different processing methods of Eucommiae Cortex can be divided into two groups by PCA analysis,and 9 different chemical constituents can be found out,and different 'sweating' methods can be divided into two groups by PCA analysis to find out the 10 different chemical constituents.6.The analysis of microbial community structure and its influence on chemical transformation during the process of 'sweating'.In this experiment,fluorescence in situ hybridization(FISH)was used to determine the samples in different stages of Eucommiae Cortex 'sweating' process.The changes of microbial community structure in different stages of'sweating' were analyzed and compared,and the endophytic strains of Eucommiae Cortex were obtained by means of isolation and purification.UFLC-Triple TOF-MS/MS was used to compare the difference from Eucommiae Cortex inoculated different strains and 'sweating' Eucommiae Cortex and sterilized Eucommiae Cortex.The results showed that the microbial community structure of Eucommiae Cortex in different stages had obvious changes,and the chemical composition of Eucommiae Cortex inoculated with endophytic bacteria was obviously different from that of dried Eucommiae Cortex and Eucommiae Cortex after 'sweating'.7.Quality analysis of different medicinal parts of Eucommia ulmoides.The chemical constituents of Eucommiae Cortex were analyzed by UFLC-Triple TOF MS/MS.UFLC-QTRAP-MS/MS was used for the simultaneous determination of 21 constituents in different parts of Eucommia ulmoides.ICP-OES was used for the simultaneous determination of 24 kinds of inorganic elements in different medicinal parts of Eucommia ulmoides.UFLC-Triple-TOF-MS/MS was used to analyse the chemical constituents and the differences of different medicinal parts of Eucommia ulmoides.The results showed that 32 chemical constituents in Eucommiae Flos Male were identified and their cleavage pathways were speculated,and there were obvious differences in chemical constituents in different medicinal parts of Eucommia ulmoides,and 23 common different constituents and their variation patterns were found out.
