Rheumatoid arthritis(RA)is an autoimmune disease with unclear etiology,chronic inflammation of joint synovium,structural destruction of joints,and malformation,and it is one of the common diseases with high morbidity.The national name of old Chinese medicine professor Chen Jipan is commonly used C.orbiculatus and S.suberectus in the treatment of rheumatoid arthritis,so that it is well known all around the nation that the preparation of Guangdong Province Chinese Medicine Hospital "Erteng Tongbi Mixture" has exact clinical effect on the joint pain,swelling and deformation of the whole body.In this study,AA rats were used to model rheumatoid arthritis to study the effects of decoction liquid obtained from C.orbiculatus and S.suberectus under different extraction conditions on the anti-rheumatoid efficacy and hepatic and renal toxicity.The phagocytic cells were used to study the effects of alcohol extracts and water extracts of C.orbiculatus and S.suberectus on cytokine secretion and antigen presentation.The results showed that the decoction of different extraction methods can all significantly improve the joint swelling of AA rats,and the alcohol extracts of this mixture had no obvious toxicity to liver and kidney of AA rats.In addition,the alcohol extracts of C.orbiculatus and S.suberectus could significantly inhibit the activation of macrophages and the secretion of proinflammatory cytokines.The alcohol extracts of C.orbiculatus and S.suberectus were extracted by different polar solvents.Then the macrophages were used to screen which part of the extracts had immunosuppressive activity.The results showed that the active ingredients of petroleum ether and dichloromethane of C,orbiculatus and the active extractions of petroleum ether and ethyl acetate of S.suberectus could significantly inhibit the activation of macrophages and the secretion of proinflammatory cytokines.The effect of Compatibility of active extractions from C.orbiculatus and S.suberectus on anti rheumatoid arthritis and reversing immune imbalance was studied by using CIA mice as the model for rheumatoid arthritis.The results showed that the active extractions remarkably inhibited joint swelling of mice,reversed the imbalance of Thl/Th2?Th17/Treg cells in spleen and lymph nodes.Also,the proinflammatory and anti-inflammatory cytokines in serum could be reversed too.In this study,the spectral response of the active extractions of C.orbiculatus was investigated.It was found that the 6th,10th,11th,12th and 13th peaks of the 13 common peaks contributed more to the reduction of IL-6 and TNF-a secretion,and the 3rd,4th,5th,7th and 9th contributed more to the increase of IL-10 secretion.The results showed that the 13th peak represented celastrine and the 6th to 11th peaks could represent sesqueteerpenoid alkaloids by mass spectrometry analysis of active extractions of C.orbiculatus.Through the determination of the content of 7 components in each part of S.suberectus,it was found that the active components were phenolic acid represented by protocatechuic acid,flavanol represented by catechin and isoflavone represented by formononetin.
