ObjectiveTo study the effect of microRNA-125a-5p on nuclear translocation of heme oxygenase 1(nuclear HO-1)in regulating hypoxia injury by establishing an in vitro hypoxic model of blood-spinal cord barrier(BSCB)When the role of the blood spinal cord barrier.MethodsThe immortalized human cerebrovascular endothelial cells(HCMEC / D3)were co-transfected with microRNA-125a-5p mimics,microRNA-125a-5p Inhibitor and HO-1C?23 HO-1C?23 + microRNA-NC group,Lv-HO-1C?23 + microRNA-125a-5p mimics group,Lv-HO-1C ? 23 group,Lv-1C ? 23 + microRNA-125a-5p Inhibitor group).In vitro BSCB model was constructed with Transwell cells in HCMEC / D3 and astrocytes.Cobalt chloride induces in vitro model of BSCB hypoxia.The expression of ZO-1,Occludin and VE-cadherin were detected by Real-time PCR and Western blot.The overflow rate of HRP was used to evaluate the permeability of BSCB in vitro and the permeability of BSCB Sex.ResultsBSCB permeability test showed that up-regulation of nuclear HO-1 could decrease the overflow rate of BSCB HRP in hypoxic injury,and on the basis of this,promote the expression of microRNA-125a-5p,the HRP spillover rate decreased again.The silencing of microRNA-125 a The expression of-5p results in an increase in HRP spillover rate.Western blot and Real-time PCR results showed that compared with the blank group,Lv-HO-1C?23 group was closely related to the adhesion-related protein and mRNA expression(P <0.05);compared with the Lv-HO-1C?23 group,Close and adhesion-related protein and mRNA expression decreased P <0.05),while enhanced histone expression increased P <0.05).ConclusionUnder hypoxic conditions,nuclear HO-1 can reduce the permeability of BSCB by upregulating microRNA-125a-5p.
