The Effect Of Yangyin Yiqi Huoxue Recipe On Tfh-related-molecules Of NOD Mice

Objective:To discuss the relationship between follicular helper T cells(Tfh)and sjogren's syndrome(S S),to study the effect of Yangyin Yiqi Huoxue Recipe(YYHP)on Tfh-related-molecules of non-obese diabetic(NOD)mice with Sjogren's Syndrome.Methods:Totally 43 NOD mice were randomly into 6 groups,i.e.the high dose Chinese medicine combined with western medicine group[administered with high dose YYHR 0,4ml and hydroxychloroquine 0.4ml by gavage],the low dose Chinese medicine combined with western medicine group[administered with low dose YYHR 0.4ml and hydroxychloroquine 0.4ml by gavage],the high dose Chinese medicine group[administered with high dose YYHR 0.4ml],the low dose Chinese medicine group[administered with low dose YYHR 0.4ml],the Western medicine group[administered with hydroxychloroquine 0.4ml by gavage],the model group.Six Balb/C mice were recruited as the normal control group.Mice were sacrificed to take submandibular gland after 8 weeks.The submandibular gland was excised.Protein expression of BCL-6 and CXCR5 in submandibular glands was detected by SP method.Results:1)There was no difference in weight,the amount of drinking water,and the results of the diet showed that the consumption of the normal group was the lowest,and it did not change with time.The model group consumes the most and increases over time.The consumption of each medication group lies between the two groups,and the high dose of Chinese medicine combined with western medicine group is the closest to the normal group.2)Submandibular gland index:There was no significant difference between Chinese and western medicine(high)group(P>0.05),but all other groups were significantly lower than normal group(P<0.05).There was no significant difference between each medication group and model group(P>0.05).The Chinese and western medicine(high)group was significantly higher than traditional Chinese medicine(low)group and hydroxychloroquine group(P<0.05).3)The pathological results of HE showed that the acinar structure was destroyed in each group of NOD mice,especially in the model group.The pathological score of submandibular gland in normal group was significantly lower than that in nod group(P<0.05),and that in traditional Chinese medicine(high)group was significantly lower than that in traditional Chinese medicine(low)group and model group(P<0.05).4)immunohistochemical:Compared with the normal group,the NOD rats were significantly higher(P<0.01).Compared with the model group,the Chinese and western medicine(high)group and the traditional Chinese medicine(high)group were significantly lower(P<0.05),but the other drug groups had no difference.Conclusion:BCL-6,CXCR5 and Tfh cells are likely to be involved in the pathogenesis of Sjogren syndrome.High dose of traditional Chinese Medicine combined with Western Medicine in the treatment of SS the best curative effect.Yangyin Yiqi Huoxue prescription can delay the progress of SS and improve the symptom treatment by regulating the expression of BCL-6 and CXCR5 and other molecules in Tfh cells.
