| Objective: Danzhi Jiangtang Capsule has efficacy observation of Yiqi Yangyin Huoxue(DJC) in type 2 diabetic patients with vascular lesions of vascular endothelial growth factor: platelet derived growth factor(PDGF) changes and v WF factor were observed and compared before and after treatment, carotid artery intima-media thickness changes, related factors and vascular endothelial type 2 diabetes mellitus endothelium injury mechanism in diabetic angiopathy, describes the traditional Chinese medicine Yiqi Yangyin Huoxue on protection of vascular endothelial injury in type 2 diabetes mellitus.Methods:1.Clinical research:2.1 Study Object:60 cases of type 2 diabetes, deficiency of Qi and Yin and blood stasis syndrome conventional oral hypoglycemic drug treatment, the treatment group in the treatment of drug control group based on the use of Danzhijiangtang capsule.Two groups were treated for 12 weeks.2.2 Study Methods:Two groups of patients before and after treatment were calculated the total efficiency and TCM syndrome integral and fasting insulin(FINS),glycosylated hemoglobin(Hb A1c), insulin resistance index(IRI), free fatty acid(FFA),total cholesterol(TC), triglyceride(TG), high density lipoprotein(HDL), low density lipoprotein(LDL), the determination of PDGF-BB, Vwf,correlation and analysis of PDGF-BB, v WF and diabetic angiopathy.2.Animal experiments:1.1 Study Object:Male Wistar rats of clean grade 60, body weight in 180g~220g, a week after adaptive feeding ordinary feed, randomly divided into 15 as the blank group(K), the other rats were given 1% of the concentration of streptozotocin(STZ) and citric acid buffer(citric acid buffer solution as 0.1mmol/L, p H4.5) injection according to the 30mg/kg body weight intraperitoneal.72 hours later, tail blood sampling,detection of blood glucose is more than 16.7mmol/L, the amount of water, food intake and urine volume increased significantly, while the success of model making.Screening and become the model of rats, which were randomly divided into model group(M),Yiqi Yangyin Huoxue Recipe of Danzhijiangtang Capsule group(Dan group, D),gliclazide group(Western medicine group, X), blank group and model group were given physiological saline, gliclazide group was treated with gliclazide gliclazide(80mg, Tid) by gavage, Danzhijiangtang Capsule group, Danzhijiangtang Capsule(3capsules each time, Tid) combined with intragastric administration of gliclazide group,each group of consecutive gavage for 8 weeks.Feeding time was 10 weeks.1.2 Study Methods:Both before and after treatment respectively before and after treatment were observed in each group in general, and the detection of FPG, 2h PG,FINS, TC, TG, HDL, LDL.For 10 weeks after the last administration, fasting for 12 hours, intraperitoneal injection of 10% chloral hydrate anesthesia, femoral artery blood,platelet derived growth factor ET-1 expression level of m RNA.Results:1.Clinical Study:Clinical observation showed that, after treatment with Danzhijiangtang Capsule the treatment group the total effective rate was 85.7%, the control group was 64.2%, good curative effect the treatment group than the control group(P<0.05); TCM syndrome integral treatment group after treatment compared with control group,were better than control group(P<0.05),compared with before treatment group therapy has improved significantly(P<0.01).After treatment, two groups of insulin resistance index were lower(P<0.01), and the treatment group was better than that of the control group(P<0.05); FFA, TG and LDL compared with those before treatment were lower(P<0.01); HDL compared with those before treatment was increased(P<0.05), and the treatment group is better than control group(P<0.01);PDGF-BB, v WF levels in patients with reduced, carotid artery intima-media thickness decreased(P<0.01).2.Animal experiments:Animal experimental results showed that,the model group,gliclazide group and Danzhijiangtang Capsule group rats after modeling, body weight, food intake and fasting blood glucose, blood lipid levels were lower than the blank group increased(P<0.01) after treatment, body weight, Danzhijiangtang Capsule food intake was lower than that of model group(P<0.01); compared with the model group, gliclazide group and Danzhijiangtang Capsule group TG levels were significantly decreased(P < 0.05 or 0.01), the HDL-C level increased significantly(P<0.01), and of Danzhijiangtang Capsule group was higher than that of gliclazide group(P<0.05); control group, modelgroup, Danzhijiangtang Capsule the rats in group ET-1m RNA expression was significantly higher than the control group(P<0.01), Danzhijiangtang Capsule group is lower than that of model group rats ET-1m RNA expression and gliclazide group(P<0.01).Conclusion:Danzhi Jiangtang Capsule can significantly improve the clinical symptoms of diabetes and vascular endothelial injury of type 2, suggesting that the mechanism may be related to the improvement of insulin resistance, decrease the expression of vascular endothelial damage factor and interfere with the oxidative stress state and anti inflammation. |
