Objective: Preliminary screening of petroleum ether,dichloromethane,n-butanol and water extract components of Mongolian Medicine Xanthoceras sorbifolia Bunge extracts on rheumatoid arthritis caused by FIA(Freund Incomplete Adjuvant).Furthermore,Mongolian medicine Xanthoceras sorbifolia Bunge extracts were used for pharmacological study on rheumatoid arthritis,so as to provide scientific basis for the anti-rheumatic effect of Xanthoceras sorbifolia Bunge.Methods:(1): 1 72 SD rats were divided into 6 groups(12 in each group).After 15 days of continuous administration,the swelling degree of the feet before and after administration was measured.2: 60 Kunming mice were randomly divided into 6 groups(10 in each group),and 15 d was administered continuously to compare the anti-inflammatory effect.The swelling and inhibition rate were calculated.The degree of swelling was expressed as the difference between the weights of the left and right ears of each rat and the inhibition rate(%)=(model group swelling degree-degree of swelling in the administration group)/model group swelling degree × 100%.(2): Exploration of experimental conditions of rheumatoid arthritis caused by FIA: SD rats were modeled with Freund's complete adjuvant(FIA)and experimental conditions were explored.(3): Study on the mechanism of anti-rheumatic arthritis in the extraction site of n-butanol from Xanthoceras sorbifolia Bunge ethanol extract: According to the above experimental conditions,70 SD rats were divided into 7 groups(12 in each group)and given continuously.After 24 days of drug use,the following indicators were measured with an enzyme-labelling device(ELASE): SOD,MDA,GSH-Px,ASO,RF,IL-1,TNF-a,UA;legs were fixed in 20% formaldehyde solution and HE was used.Staining method to observe the pathological changes of synovial tissue.(4)HPLC method was used to compare the fingerprints of ethanol extracts from Xanthoceras sorbifolia Bunge with different solvents.The chromatographic conditions were ODSC18 column(4.6 x 300 mm,5 m),acetonitrile(B)and 0.1% acetic acid(A)as the mobile phase.The detection wavelength was 274 nm,gradient elution,flow rate 1mL/min,sample volume 5 L,column temperature 30°C.Results:(1)From the above experimental results,the n-butanol extract of Xanthoceras sorbifolia Bunge had significant anti-inflammatory effects,with significant differences(P<0.05).(2)The pharmacological study of anti-rheumatoid arthritis in the anti-inflammatory parts of the ethanol extract of the Xanthoceras sorbifolia Bunge was further studied.The results showed that the activity of SOD,MDA,GSH-Px,ASO,RF,IL-1,TNF-a and UA in the high dose group of n-butanol extraction group was significantly different(P < 0.05).The activity of serum MDA,GSH-Px,ASO,RF,IL-1,TNF-a and UA in the small dose group of n-butanol extract was significantly different(P < 0.05).The activities of SOD,ASO,RF,IL-1 and TNF-a in serum of SD rats were significantly different(P < 0.05).Conclusion: N-butanol extract from ethanol extract of Xanthoceras sorbifolia Bunge has obvious anti-rheumatic effects.
