Objective:To investigate PRL-3 gene promotes the invasion and metastasis of peritoneum of gastric cancer whether through PI3K/AKT signaling pathway in SGC7901 cells and nude mice.Methods:In our previous study,two normal gastric epithelial cell lines(GES-1 and HFE-145)and four different gastric cancer cell lines(MKN28,SGC7901,MKN45,BGC823)were cultured and the PRL-3 protein level was detected by Western Blot.We found that the PRL-3 protein level in SGC7901 cell line was higher than others.Then,the EGFP-PRL-3 plasmid was constructed and transfected into SGC7901 cell line.The recombinant empty plasmid was transfected into SGC7901 cell line.The concentration of LY294002(0uM,5uM,10 uM,20uM,40uM)were detected by CCK-8 method.After treatment with LY294002 inhibitor,the expression of PRL-3,AKT,p-AKT,MMP-2,MMP-9 protein and invasion and metastasis of cells in each group were detected by western blot and Transwell invasion assay.We established the nude mouse peritoneal metastasis model by intraperitoneal injection.The mice were sacrificed by cervical dislocation and the peritoneal cavity was opened for visual inspection and photography after 4 weeks.Results:The expression of p-AKT,MMP-2 and MMP-9 was significantly inhibited by PRL-3/SGC7901 in PI3 K inhibitor LY294002.Transwell invasion assay showed that gastric cancer PRL-3/SGC7901 treated with inhibitor LY294002,Invasion and migration capacity were significantly decreased;in animal experiments,plus the inhibitor group more than 1mm tumor number was significantly less than the control group and the difference was statistically significant.Conclusion:PRL-3 gene may be through PI3K/AKT signaling pathway to promote the invasion and metastasis of peritoneum in gastric cancer cells and nude mice.
