PART ? The Characteristics of acute infection of Pseudomonas aeruginosa in the intraperitoneal cavity of mouseObjective To establish a mouse model of Pseudomonas aeruginosa(P.a)early biofilm(BF)infection in the intraperitoneal cavity of mouse.The characteristics of P.a infection in the intraperitoneal cavity of mouse were observed.Method(1)Establishment of P.a biofilm in vitro: Disposable infusion tube that made by PVC was used as the implant and PAO1 wild strains as the experimental strains.The model of intraperitoneal implant BF infection was established.(2)Establishment of P.a infection model in the intraperitoneal cavity of mouse.14 female mice were randomly assigned to 2 groups with 7 in each: uninfective vehicle control groups(Sterile carrier),P.a group(carrier with P.a biofilm).(3)Specimen collection: To observe pathological changes with the HE staining and the inflammation response of the local peritoneal tissue in mice.The number of white blood cells in peripheral blood of each group was counted. Remove the implants from the peritoneal cavity after 24 h and before implant.Samples of implants were counted with the continuous dilution method.The BF morphological changes were observed on the implant surface by scanning electron microscope(SEM).Results The P.a group showed obvious congestion around the plant,control group without inflammatory congestion change,only slightly wrapped phenomenon after 24 hours.The pathology changes of intraperitoneal tissue in the P.a group showed obviously infiltrated with neutrophil.The control group only observed slightly inflammation.The white blood cell count in P.a group was significantly higher than that in the control group after 24h(P<0.05).Bacterial counts of the P.a group after 24 h on the implant surface were significantly higher than the group before 24h(P<0.01).The bacterial adhesion and the biofilm formation of the group before 24 h on the implant surface was more thicker than the P.a group on SEM.Conclusion Establishment of Pseudomonas aeruginosa infection model in the intraperitoneal cavity of mouse is successful by implant of PVC material.The infective mice showed a significant high in the white blood cell count.The pathology changes of intraperitoneal tissue showed obviously infiltrated with neutrophil and lymphocytes.PART ? The effects of baicalein on Pseudomonas aeruginosa biofilm infection in vivoObjective(1)To establish a mouse model of Pseudomonas aeruginosa(P.a)early biofilm(BF)infection in the intraperitoneal cavity of mouse.(2)To investigate the therapeutic effect of baicalein on BF infection in the intraperitoneal cavity of mouse.Method(1)Establishment of P.a biofilm in vitro: Disposable infusion tube that made by PVC was used as the implant and PAO1 wild strains as the experimental strains.The model of intraperitoneal implant BF infection was established.(2)14 female mice were randomly assigned to 2 groups with 7 in each: the control group,the BCA group(300mg/kg.d).(3)Establishment of P.a infection model in the intraperitoneal cavity of mouse.Mouse in the BCA group were treated with intragastric administration of BCA;Mouse in the control group were treated with intragastric administration of sodium carboxymethyl cellulose;(4)Specimen collection: The number of white blood cells in peripheral blood of each group was counted.The concentration of CRP,TNF-?and IL-1 was measured with enzyme-linked immunosorbent assay(ELISA).Remove the implants from the peritoneal cavity after 24 h.Samples of implants were counted with the continuous dilution method.The BF morphological changes were observed on the implant surface by scanning electron microscope(SEM).The inflammation response of the local peritoneal tissue was observed.Results The white blood cell count in the BCA group was significantly less than that in the control group after 24h(P<0.05).CRP,TNF-?and IL-1 level in the BCA group was significantly less than that in the control group after 24h(P<0.01).Bacterial counts of the BCA group after 24 h on the implant surface were significantly less than the control group(P<0.01).The bacterial adhesion and the biofilm formation of the BCA group on the implant surface was more thicker than the control group on SEM.The pathology changes of intraperitoneal tissue in the control group showed obviously infiltrated with neutrophil and lymphocyte.The BCA group only observed slightly inflammation.Conclusion Baicalein can inhibit peritoneal biofilm of P.a in mice,which can significantly improve the inflammatory response.
