Objective:To investigate the protective effect of sulforaphane?SULFORAPHANE,SFN?on oxidative damage of human umbilical vein endothelial cells?HUVEC?induced by hydrogen peroxide?H2O2?,and to further study the mechanism of sulforaphane in the protection of endothelial cell injury.Methods:The human umbilical vein endothelial cell line provided by Shanghai cell bank of Chinese Academy of Sciences was resuscitated,and the cells were cultured at 37 5%CO2and over 80%degree of fusion.Experimental groups:?1?Blank control group?Control group?:The cells were cultured for 2.5 hours.?2?Hydrogen peroxide group?group H2O2?:To give separately 100.200.400.600.800?M cells were cultured for 2 hours,and 400?M was selected as the best intervention concentration.?If the following groups are not specified,the default H2O2 concentration is 400?M?;?3?Sulforaphane+hydrogen peroxide group?group SFN+H2O2?:Different concentration of SFN2.5.5.10?M)after incubating for 30 min,2h was continued to be cultured with 400?M H2O2.To collect the cells after intervention,the expression of intracellular transcription factor E2?NRF2?mRNA and heme oxygenase-1?HO-1?mRNA was determined by using MTT assay to compare cell activity,flow cytometry assay of intracellular ROS levels,and reverse transcriptase polymerase chain reaction?RT-PCR?.Results:Compared with the control group,the cell absorbance value?a value?of H2O2 group was significantly reduced?p<0.05?,and the value of a was decreased by the concentration dependence,and the absorbance value of SFN+H2O2 group was higher than that of H2O2Group?p<0.05?.The level of ROS in H2O2 group was higher than that in control group?p<0.05?,and the ROS level in SFN+H2O2 group was lower than that in H2O2 Group?p<0.05?.The expression level of Nrf2 mRNA and HO-1 mRNA in H2O2 group was lower than that in control group?p<0.05?,and HO-1 m RNA and Nrf2 mRNA expression in SFN+H2O2 group were higher than that in H2O2 Group?p<0.05?.Conclusion:Sulforaphane has protective effect on oxidative damage of HUVEC caused by hydro-gen peroxide,and its mechanism may be related to the expression of sulforaphane in trans-cription level activation Nrf2/HO-1.
