Objective Discussed by using modified line plug,rabbit as the subjects of focal cerebral ischemia model.Methods Using the random number table method,Before building 45 healthy clean level only adult New Zealand rabbits were divided into control group(5)and model group(40),Dip in with homemade head end with paraffin 2-0 fishing line as the line switch,By residual snip,external carotid artery after insert the intracranial internal carotid artery,to the congestion in the brain artery,After 6 h to nerve function score,Nerve function defect grade 2 or more divided into building success.Anesthesia executed rabbit,With 2% 2,3,5-triphenyl tetrazolium chloride solution to the dyeing of slices,observe the infarcts.In model group building success,failure,and death experimental rabbit line bolt head end diameter,thread bolt insertion depth contrast.Results In the 40 model group animals,six of them died,Including postoperative 4 h within four died in subarachnoid hemorrhage;there were 2 animals dead of anesthesia accident,The mortality was 15.0%;There were 7 animals failed,Mainly because of cerebral vasospasm and inadequate line bolt insertion depth;Building successful 27 animals,the success rate is 67.5%.In the control group,all rabbits lived.Building successful line bolt head end diameter of rabbit,insert depth,respectively,compared with death and the outcome of the rabbit,differences were statistically significant[diameter(mm):(0.52 ±0.14)than(0.45 ± 0.40)and(0.58 ±0.17);the depth(cm):(5.49 ± 0.17)than(6.04 ± 0.11)and(4.26 ± 0.30);P < 0.05).Conclusions Improved line plug method can be successful in the preparation of rabbit brain artery focal cerebral ischemia model,simple operation,good repeatability and practicability.Objective This study through the detection of after hyperbaric oxygen exposure,the expression of internal carotid artery and vertebral basilar artery ET-1 and eNOS and their different receptor subtype to explore the hyperbaric oxygen under the action of endothelin 1(ET-1)and endothelial nitric oxide synthase(eNOS)and its receptor subtype expression changes.And through the suture method to establish the rabbits middle cerebral artery occlusion cerebral ischemia ischemia model.Further clarify the mechanism of hyperbaric oxygen exposure cause anterior circulation shrink and posterior circulation expand,and in this way to compare the different influence between Cerebral infarction individual and normal individual under the effect of hyperbaric oxygen on brain circulation regulation.Methods 40 healthy adult New Zealand rabbits whether they are Male and female were randomly divided into 2 groups:the surgical group and the control group,The number of each group is 10 and 30 respectively.The rabbits in the surgical group then divided into cerebral ischemia(n = 11)and hyperbaric oxygen cerebral ischemia group(n = 11),The remaining 8 rabbits were excluded because of death or neurological function score failed to reach the standard.The rabbits in the control group only separation neck vascular and then suture neck incision the rest do not do any processing.The establishment of cerebral ischemia model by the method of thread embolus in the operation group.hyperbaric oxygen cerebral ischemia group were exposed to HBO at 2.2ATA for 60 minutes for 3 days,The rabbits in the control group and ischemic group animals not hyperbaric oxygen absorption,live for three days.All animals were killed 3 days after the use of ET-1,ETAR and eNOS antibodies and then immunohistochemical staining was carried out to observe the expression of ET-1,ETAR and eNOS in the 3 groups of internal carotid artery and basilar artery.Results ET-1,eNOS and ETAR were expressed in the internal carotid artery and basilar artery of the 3 groups of rabbits.Compared with the sham operation group and the cerebral ischemia group,the expression of ET-1 and ETAR receptors in the internal carotid artery increased significantly,and the expression of eNOS in the basilar artery increased significantly compared with the internal carotid artery.Conclusion HBO exposure can enhance the expression of vasoconstrictor factor ET-1 and ETAR in the internal carotid artery of rabbits with cerebral ischemia,the expression of basilar vascular factor eNOS is enhanced and the anterior circulation systolic circulation expands.Hyperbaric oxygen has the same effect on normal rabbits and cerebral ischemia model rabbits.
