Objective: The purpose of this study was to investigate the recovery mechanism of mild-pressure hyperbaric therapy after high intensity training around oxidative stress and capillary regenaration.We discussed the production and cleanup of free radical,the gene and protein expression of capillary regeneration related factors.So we will provide theoretical basis and experiments support for the application of mild-pressure hyperbaric therapy to recovery after training.Method: Thirty male SD rats(8 week old)were selected by adaptability training from forty rats,and they were randomly divided into three groups:control group(C),quiet rest after high intensity training(HQ,28m/min speed of 10°slope),mild-pressure hyperbaric therapy after high intensity traininng(HM)with ten rats in each group.Oxygen concentration of mild-pressure hyperbaric therapy was about 26%,and the pressure was 3.6PSI.The rats were trained 1h/d,5d/week for 8week.The rats of HM immediately received Mild-pressure hyperbaric therapy for 1h after training.After 8week's training,at the 24 h after last training they were sacrificed.The left gastrocnemius was cut to detect oxidative stress factor and observe the m o rp hol o g y s t ru ctu re.R T-PCR,W es t ern Bl ot were us ed t o d et ec t VEGF,VEGFR-2,b FGF m RNA and protein expression of rat gastrocnemius.SPSS19.0 was used to process data and P<0.05 has significant difference.Results: 1.Changes in time to exhausion: the time to exhausition of HM group was significantly longer than HQ group(P<0.05);2.Changes in oxidative stress parameters: the The MDA level of HQ group was significantly higer than C group and HM group(P<0.05).SOD activity of HM group was significantly higher than C group(P<0.05).GSH-PX activity of HM group was higer than C group and HQ group,and there was significantly difference(P<0.05).3.The morphology structure changes of skeletal muscle: Compared to C group,muscle injury of HM group and HQ group was more serious,but HM group was less serious than HQ group.4.The effect of Mild-pressure hyperbaric therapy on microvascular vascular density and capillary regeneration related factors :4.1 The expression of capillary regeneration related factors: The VEGF protein expression of HQ group was higher than C group and HM group(P<0.05).VEGFR2 m RNA expression of HQ group was higher than HM group(P<0.05).The VEGFR2 protein expression of HQ group was higher than C group and HM group(P<0.05).b FGFm RNA expression of HM group was lower than HQ group and has significantly difference(P<0.05).b FGF protein expression of HQ group was higher than C group and HM group,and has significantly difference(P<0.05).4.2 Microvascular vascular density:MVD of HM group was higher than C group,and has very significantly difference(P<0.05).HM group was higher than HQ group and has significantly difference(P<0.05).HQ group was higher than C group and has significantly difference.Conclusion: 1.High intensity training can lead to skeletal muscle oxidative stress injury and has poor sport performance.Mild-pressure hyperbaric therapy can improve the activity of antioxidant enzymes of skeletal muscle and reduce the lipid peroxidation,thus improve exercise capacity.2.High intensity training can cause MVD increse of skeletal muscle.Mild-pressure hyperbaric therapy can accelerate the generation of capillaries and promote the transport of nutrients,oxygen and metabolites,therefore can be helpful in the recovery process.
