| Objective:The regulation on CX43 GSK3β and Bcl-2 of VPA autism model rats after swimming and enriched environment.Methods:One of the female and male Wistar rats were placed in the same cage the whole night,and when the yellowish or white translucent shade was kept in the vagina,the females were removed from the plate,which was recorded for the first day.The pregnant rats were sacrificed separately and the pregnant females were randomly divided into two groups.One group was treated with 0.85% saline and VPA powder which was made into 250 mg / ml solution.The solution was incubated at 12.5 days(600mg / kg),the offspring of the pregnant group was recorded as the model group,the control group was not any intervention.In this paper,the experiment is divided into five groups: swimming + rich environment group,rich environment group,autism group,swimming group,blank control group.The swimming group was given moderate exercise training for 90 minutes in water at 30 ± 1 ° C every afternoon at 3:00-4: 30 and continued training for 8 weeks.Enriched environment group living in a long,wide,high 1m × 0.6m × 0.4m transparent cage,this cage with a variety of toys,so that autistic rats can be trained.At the end of the intervention,the expression of CX43,GSK3β and Bcl-2 protein in the cerebral cortex of rats were detected by immunohistochemical method.The image-based analysis was carried out by using Image-Pro Plus multifunctional true color image analysis and management system.The fluorescence intensity(fluorescence intensity / total area)of CX43,GSK3β,Bcl-2 protein was calculated and compared.The experimental data were analyzed by SPSS13.0 statistical software.Experimental results:1.Comparison of body weight: VPA autism model rats and normal rats compared to the weight of 7 days after birth,14 days,21 days,PN7 <0.05,there was a significant difference;PN14 and PN21 were> 0.05,No significant difference.2.The eye opening time of PN13,PN14 and PN15 were significantly higher in the VPA model group than in the normal group(P <0.05).3.Directional trend: Compared with the normal group of young rats,autistic young mice turned 180o longer,in PN7,PN8 and PN9 days,P <0.05,with statistical differences.4.Forced swimming: PN8,PN10 days,autistic model rats swimming ability were significantly lower than normal rats,P ≤ 0.05.5.Compared with the model group,the CX43 protein expression in the cortical cortex of the swimming and enriched environment group was significantly higher than that in the model group(P <0.05);Compared with the swimming group,the CX43 protein expression in the rich environment group was higher than that in the swimming group <0.05.6.The expression of GSK3β protein in the swimming and enriched environment group was significantly higher than that in the model group(P <0.05),and the expression of GSK3β protein in swimming group was significantly higher than that in the enriched environment group(P < 0.05,with significant differences.7.The expression of Bcl-2 protein in the cerebral cortex of the swimming and enriched environment group was significantly lower than that of the model group(P<0.05),and the expression of Bcl-2 protein in the enriched environment group was similar with that in the swimming group P> 0.05,without significant differences.Inconclusion:Enriched environment and swimming can increase the expression of CX43 and GSK3β protein in the cerebral cortex of autistic model rats and decrease the expression of Bcl-2 protein. |
