The functional of intestinal barrier can be changed by prolonged exhausting exercise.Intestinal mucous mechanical barrier is the first defense in the intestinal barrier.The integrity of mucous mechanical barrier function is the safeguard of the other barriers function.This research used treadmill for 9 months prolonged exhausting exercise to build model and aim to find the improving effect of lactobacillus rhamnosus on mucous mechanical barrier after prolonged exhausting exercise.Methods:The 40 experimental rats randomly divided into 4groups.8 rats in control group,12 rats exercise group,8 rats probiotics groupand 12 rats in exercise&probiotics group.Exercise group and exercise&probiotics group were both exercised to exhaustion on treadmill for 9month.Exercise content:The first week,10m/min×30min adaptive training everyday?From the second week to 9th week,exhaustive training everyday,Sunday rest.Exercise start at 10m/min,increasing 5m/min speed in every 5 minutes till 40m/min.The exercise be terminated when the rats exhaustion.The standard of the exhaustion is that rats lie on its stomach and cannot moving with stimulation of light and electric shock.Probiotics group and exercise&probiotics group were given a gavage of with Lactobacillus rhamnosus GG 2ml in dosage of 107CFU/m L once a day.The other groups were given 2m L distilled water.The rats were killed by 2m L nembutal in 2% dosage 24 hours after the last time exercise.Collect blood from aorta abdominalis.Cut out 2cm jejunum from near-end intestine 5cm place.Cut out 2cm jejunum and put it into formaldehyde solution.The protein content of occludin and zonulin was detected by western blot.The content of serum LPS was detected by radio-immunity.Assess intestinal morphology by histochemical.Results:Compared with the control group,content of serum LPS obviously increased in probiotics group(P <0.05).Serum LPS content in exercise&probiotics group is obviously less than exercise group(P <0.05).Intestinal villus no edema and lamina propria no damaged in control group.Edema intestinal villus and damaged lamina propria can be found in exercise group.Edema intestinal villus also can be found in probiotics group and exercise&probiotics group.But no damaged lamina propria in this two group.Compared with the control group,the protein content of occludin is obviously less in exercise group.There is no obviously difference of zonulin protein content in every group.Conclusion: 1 The functional of intestinal barrier can be weakened by Prolonged exhausting exercise.The functional of intestinal barrier can be improved by lactobacillus rhamnosus and prevent the increasing of serum LPS.Using single kind of probiotics can increase the content of serum LPS in normal condition.2 Intestinal villus can be edema and lamina propria can be damaged by Prolonged exhausting exercise?The damaged of lamina propria can be amelioration by lactobacillus rhamnosus.But lactobacillus rhamnosus is no effect on Intestinal villus edema.3 The protein content of occludin was decreased by Prolonged exhausting exercise but no effect on zonulin content.There is no effect on increasing the occludin content after Prolonged exhausting exercise by using lactobacillus rhamnosus.
