Objective: To observe the effect of heavy load on the expression of JNK,IRE1,TRAF2 and ASK1 in skeletal muscle cells after exercise in skeletal muscle of rats and the effects of IRE1-JNK Channel in the role of endoplasmic reticulum stress in skeletal muscle cell apoptosis.Methods: Male healthy 8-week-old rats were randomly divided into quiet control group,exercise group.The movement plan of the exercise group is continuous downhill running,the runway gradient is-16 °,the speed is 16 m / min,the time is 90 min.According to the time points,the rats were divided into 0h group,12 h group,24 h group,48 h group and 72 h group after exercise.The expression of GRP78,JNK,IRE1,TRAF2 and ASK1 were detected by western blot.The gray value of the corresponding bands was analyzed by Gel-pro software.The results of single factor analysis of variance were used to analyze the expression of Caspase3.Processing data.The activity of Caspase3 in the soleus muscle reached its peak at 12 h after heavy load exercise.Compared with the quiet control group,there was no significant difference between the 72 h group and the quiet group(P> 0.05)The activity of Caspase3 in soleus muscle was significantly higher than that in quiet group(P <0.05).The expression of GRP78 protein in the soleus muscle reached the peak at 12 h after heavy load exercise.Compared with the quiet control group,the concentration of GRP78 protein in the soleus muscle was significantly higher than that in the quiet group(P <0.05)<0.05).The expression of IRE1,TRAF2,ASK1 and JNK protein in the soleus muscle reached the peak at 12 h after heavy load exercise.Compared with the quiet control group,IRE1,TRAF2,ASK1 and JNK protein were expressed in the soleus muscle after heavy load exercise The concentration of phase was significantly higher than that of quiet group(P <0.05).Conclusion: A large-load exercise can lead to skeletal muscle cell apoptosis,which may be due to exercise-induced endoplasmic reticulum stress,activation of IRE1-JNK pathway.
