Objective:To Observe the influence of intensive exercise on endoplasmic reticulum stress in rat skeletal muscle,and a heavy load exercise after GRP78,PERK,p-eIF2?,ATF4 protein expression,while giving specific inhibitors,observe the expression change of PERK pathway protein,explore the PERK pathway in exercise induced endoplasmic reticulum stress in skeletal muscle effect.Methods: A total of 8-week-old male SD rats were randomly divided into C,E,A,Z and ZE five groups.Among them,the E group,A group,Z group,ZE group were divided into 0h,12 h,24h,48 h and 72 h,a total of 21 groups,8 rats in each group.The scheme adopted by the sports group is a one-off downhill running,the slope of the running platform is-16°,and the time is 90 min.In the A group,the rats were given a 0.5% DMSO solution,which was dissolved in the saline solution.After gavage,30 min began to timed.A specific inhibitor GSK2606414(1.0mg/kg)was given to the Z group and the ZE group,at the time of 30 min was immediately after gavage.In the solution,the specific inhibitor was first dissolved in the corresponding DMSO solution with a final concentration of 0.5%,and then diluted with saline to the target concentration.After taking exercise or gavage,the soleus muscle of rats was measured at corresponding time phase.The protein expression of GRP78,PERK,p-eIF2? and ATF4 was detected by Western blot.The corresponding gray value of strips was analyzed by Gel-pro software,and processed by SPSS One Way ANOVA.Results: 1.GRP78 rat soleus muscle after exercise in protein increased significantly after exercise,0h,12 h,24h remained at a high level,and the C group showed significant difference(p<0.05);PERK,p-eIF2?,ATF4 protein expression increased significantly(p<0.05),12 h,24h remains at a high level;2.After applied to specific inhibitors,GRP78 protein expression compared to the exercise group significantly reduced(p<0.05);the expression of PERK,p-eIF2? and ATF4 protein are also reduced.Conclusion: 1.A heavy load exercise can induce endoplasmic reticulum stress and activate the PERK pathway;2.,after giving specific inhibitors,the PERK pathway is blocked and the endoplasmic reticulum stress decreases,suggesting that PERK pathway is one of the mechanisms of exercise induced ERS.
